As an important means of disease analysis,immuassay technology has always been the main contents of life science research.Immunodiagnostic techniques with low cost,simple operation and accurate results have always been the direction of researchers and industrial circles.The immuassay technology based on microfluidic chip has the advantages of short analysis time,high sensitivity and high-throughput detection,but it is difficult to popularize because of its difficulty in production and high cost.The difficulty of promoting the practical application of microfluidic chip is how to realize the manufacture and use of microfluidic chip at a lower cost.In this paper,a novel microfluidic chip based on capillary was developed to detect tumor markers and myocardial markers in blood samples.This paper mainly studies from the following two aspects:(1)In order to realize rapid detection of tumor markers in vitro,a micro fluidic chip based on chemiluminescence was successfully developed by immobilized primary antiprobe on the inner wall of glass capillary,which can realize rapid detection of tumor markers.The steps of surface modification,probe immobilization and immunoreaction were optimized.The chip was used to quantitatively detect the blood samples of cancer patients and the results are accurate.In addition,a zonal fixation technique was used to immobilize a variety of primary anti-probe on the inner wall of the microchannel.And the detection of multi-index was demonstrated.(2)In order to overcome the problem of high performance and cost of detection instruments for chemiluminescence detection,two platforms of fluorescence detection were designed and set up to detect the results of laser-induced fluorescence.The parameters and performance of the testing platform were optimized and investigated,and the testing platform met the testing requirements.Surface modification and longterm preservation of microchannels were also optimized.A system based on laser induced fluorescence was successfully developed for the detection of myocardial markers,and the myocardial markers in actual blood samples were detected.The results were compared with those of full-automatic chemiluminescence and immunochromatography.The microfluidic chip and its detection platform developed in this paper can realize fast,accurate and low-cost detection for the test samples.The microfluidic chip in this paper didn’t need micromachining processes,was easy to process,and had low cost,which broadened the idea of immuassay and had significant significance and value for promoting the marketing of microfluidic chips.
