Objective: The purpose of this study was to induce the establishment of functional dyspepsia(FD)rat model,and treated model with intraperitoneal injection of paeoniflorin,then explored the effect of paeoniflorin on FD rat,and clarified the relevant mechanism.Methods: In this experiment,we used iodoacetamide(IA)to induce the IA-FD rat model and clonidine(CD)to induce the CD-FD rat model.The models were tested for gastric compliance,gastric sensitivity and gastric emptying rate to evaluate the success of the animal model.Acetyl cholinesterase(Ach E)in hypothalamus and stomach tissues and serum acetylcholine(Ach)of rats were detected.The protein expression of cyclooxygenase-2(COX-2)in rat gastric tissues was detected by immunofluorescence(IF),and the Occludin,ZO-1 and E-cadherin expression of rats was detected by western blot(WB).Results: 1.Gastric compliance test: In the IA-FD model and the CD-FD model,under20 mm Hg pressure,there was no significant difference in gastric compliance between the sham group and the treatment groups(P> 0.05);under 40 mm Hg-80mm Hg pressure,compared with the Sham group,gastric compliance in the model group was significantly reduced(P <0.01);compared with the model group,gastric compliance in the different concentrations of paeoniflorin treatment groups was significantly increased(P <0.05).2.Gastric sensitivity test: In the IA-FD model and the CD-FD model,compared with the Sham group,the gastric sensitivity of the model group was significantly increased(P <0.01).Compared with the model group,the gastric sensitivity of the treatment with paeoniflorin group was significantly reduced(P <0.05).3.Gastric emptying rate experiment: In the IA-FD model,the gastric emptying rate of the model group is significantly different from that of the Sham group.The model group is significantly lower than the Sham group(P <0.01).Compared with the model group,IA-FD + 15 mg / kg / d Pae treatment group increased the gastric emptying rate(P <0.05).In the CD-FD model,compared with the Sham group,the gastric emptying rate in the model group was reduced(P <0.01),which has a significant difference.Compared with the CD-FD model group,the gastric emptying rate in different concentrations of the paeoniflorin treatment group all increased(P <0.05).4.Ach activity in serum: In the IA-FD model,compared with the Sham group,the Ach activity in the serum of the model group was significantly reduced(P <0.01).Compared with the model group,Ach activity was significantly increased in the serum of IA-FD + 30 mg / kg / d Pae treatment group rats(P <0.05).In the CD-FD model,compared with the Sham group,the Ach activity in the serum of the model group was significantly reduced(P <0.01).Compared with the model group,the Ach activity of the paeoniflorin treatment group at different concentrations was significantly increased(P < 0.05).5.Ach E activity in the hypothalamus and stomach: In the IA-FD model,there was no significant difference of in each group of rats(P> 0.05).In the CD-FD model,compared with the Sham group,the Ach E activity in the hypothalamus and stomach of the model group rats was increased(P <0.01).Compared with the model group,the Ach E activity in different concentrations of paeoniflorin-treated groups were decreased(P <0.05).6.Immunofluorescence: In the IA-FD model and the CD-FD model,the expression of COX-2 in the Sham group was small.Compared with the Sham group,the expression of COX-2 in the model group was significantly increased;compared with the model group,the expression of COX-2 in the paeoniflorin treatment groups were significantly reduced.7.Western blot: In the IA-FD model and the CD-FD model,compared with the Sham group,the expression of Occludin protein in the duodenum of the model group was significantly reduced(P <0.01);compared with the model group,the expression of Occludin protein in the 30 mg/kg/d Pae treatment group increased(P <0.05).In both IA-FD and CD-FD models,compared with the Sham group,the expression of ZO-1protein in the duodenum of the model group was significantly reduced(P <0.01),compared with the model group,the expression of ZO-1 protein in the 30 mg / kg / d Pae treatment group increased significantly(P <0.05).Conclusion: Paeoniflorin can improve gastric emptying rate and gastric compliance,reduce gastric sensitivity in FD rats,and thereby improve gastric motility disorders in FD rats.Paeoniflorin may increase Ach activity,down-regulate Ach E activity,and then promote gastrointestinal motility to treat FD.Paeoniflorin may play a therapeutic role in FD by improving inflammation in the stomach and enhancing the integrity of the intestinal mucosa of FD rats.
