Objective:(1)To study the pathological pictures of gastrointestinal mucosa in rats with gastrointestinal mucosal injury,as well as the number of writhing and open field experiments in rats.After the rats with gastrointestinal mucosal injury were intervened by rubbing their abdomen,the gastrointestinal mucosal injury and the changes in the number of writhing and open field test scores were observed.To verify that the absolute ethanol gavage method can create a gastrointestinal mucosal injury rat model,and the rubbing method can effectively improve the gastrointestinal mucosal condition in rats with gastrointestinal mucosal injury.(2)The study is to observe the morphological changes of the gastric mucosa of rats in different groups,and detect the levels of ITFm RNA in the hypothalamus and the content of ITF in the serum to explore the possible mechanism of rubbing abdomen on rats with gastric mucosal injury.Methods:(1)Experiment 1: Select 30 male healthy SD rats.First carry out adaptive feeding for a week.Thirty rats were randomly divided into two groups: normal control group and model group,of which 10 were normal group and 20 were model group.The rat model of gastrointestinal mucosal injury was made by intragastric administration of absolute ethanol.The SD rats with gastrointestinal mucosal injury that have been successfully modeled were randomly divided into abdominal massage group and model control group,with 10 rats in each group.Abdominal massage group: model building,with abdomen rubbing operation,normal food and water every day;model control group: model building,no intervention,normal feed and water every day;normal control group: no model building And abdominal massage,normal feeding and water daily.After modeling and rubbing the abdomen intervention,the rats' gastric tissue damage was observed by naked eyes,and the behavioral changes of rats in each group were detected by writhing experiment and open field experiment.(2)Experiment 2: The method of modeling and grouping is as experiment 1.Laboratory testing is carried out after successful model building.Take the hypothalamic tissue,blood and stomach tissue of three groups of rats.Detect the expression level of ITFm RNA in hypothalamus and the content of ITF in blood,and observe the morphological changes of stomach tissue and the changes of inflammatory factors in the gastrointestinal area.Results:(1)The rats in the normal control group did not have obvious writhing phenomenon,and the horizontal and vertical scores of the open field experiment were lower than those of the modeled two groups.The writhing response of the two groups of rats was obvious,and there was no statistical difference in the number of writhing responses and the scores in the open field experiment.After the intervention of rubbing the abdomen,the number of writhing and open field scores in the abdominal massage group decreased.Compared with the model control group,the difference was statistically significant(P<0.05).(2)The gastrointestinal mucosa tissues of the three groups of rats were observed by naked eyes and HE staining,and the secretion of inflammatory factors at the injured site was analyzed.The gastrointestinal mucosal injury in the two groups was obvious,and the inflammatory reaction was increased;the gastrointestinal mucosal injury in the abdominal massage group after rubbing the abdomen was less than that in the model control group,the bleeding points in the gastrointestinal mucosa were reduced,the mucosal folds were relatively intact and inflammatory Factor secretion is down-regulated.In the model two groups,the expression of ITFm RNA in hypothalamus and serum ITF in the abdominal massage group increased after intervention,which was statistically different from the model control group.Conclusion:(1)Anhydrous ethanol gavage method can successfully replicate the gastrointestinal mucosal injury rat model,and the intervention of rubbing abdomen method can significantly improve its behavioral performance.(2)The rubbing method can use the brain-gut axis pathway to upload gastrointestinal mucosal injury information to the hypothalamus,and the hypothalamus releases ITF factors to the gastrointestinal mucosa to regulate injury repair.
