Background and purpose:No matter in China or western countries,breast cancer is the most common tumor among women,and its morbidity and mortality are still on the rise.Surgery is still the most current one of the main treatment option for patients with breast cancer,whether perioperative medication will affect the recurrence and metastasis of tumors and promote the development of tumors has attracted widespread attention.Therefore,in order to explore this issue,Whether dexmedetomidine(DEX)affects the proliferation,migration and invasion ability of triple-negative breast cancer cells HS-578 T is investigated to provide a theoretical basis for preventing recurrence and metastasis of breast cancer.Methods:The HS-578 T stable cell line model of triple negative breast cancer cells was established.The experiment consists of a control group,5?mol/L,1 ?mol/L,0.1 ?mol/L,0.01 ?mol/L dexmedetomidine group.CCK8 assay was used to detect the changes of proliferation of HS-578 T breast cancer cells treated with dexmedetomidine at differentconcentrations for 0?24?48h;Wound healing assay was used to detect the change of the number of dexmedetomidine in HS-578 T breast cancer cells at different concentrations after 0,6,12,24 h;Transwell invasion assay was used to detect the number of cells invading into the lower compartment of HS-578 T breast cancer cells treated with dexmedetomidine at different concentrations for 24 h.Results:The HS-578 T model of triple negative breast cancer cell line was successfully constructed.CCK8 experimental results showed that compared with the control group,there was no statistically significant difference in OD value after dexmedetomidine was applied to HS-578 T breast cancer cells for 0?24?48 hours(p >0.05).The results of Wound healing assay showed that the number of migration cells in dexmedetomidine group increased significantly(p < 0.05),compared with the control group(118.4±10.762),0.01 ?mol/L(137±17.492),0.1?mol/L(143.2±12.056),and 1 ?mol/L(147±7.694).Transwell invasion experiment showed that,compared with the control group,after dexmedetomidine was applied to HS-578 T breast cancer cells for 24 h,the number of cells crossing the compartment membrane in dexmedetomidine group(243.6±51.58),0.1?mol/L(227.07±48.38),and1?mol/L(224.33±26.52)significantly increased compared with thecontrol group(145.08±33.96),with statistically significant differences(p< 0.05).Conclusion:Dexmedetomidine has no effect on the proliferation of HS-578 T triple negative breast cancer cells,but can increase the invasion and migration of HS-578 T triple negative breast cancer cells.
