Expression Of ROR1 In Triple-negative Breast Cancer And Its Prognostic Significance

Objective:To investigate the expression level of the receptor tyrosine kinase like orphan receptor 1(ROR1)in triple-negative breast cancer,and analyze its relationship with clinicopathological features and prognosis,so as to provide more clinical basis for future ROR1 targeted therapy.Methods:We collected the clinical data of 370 patients with breast cancer in Yan'an University affiliated hospital from January 2013 to January 2017.The 370 patients had complete clinical and pathological data.The patients did not receive neoadjuvant chemotherapy and did not have distant metastasis.Among them,69 cases of triple-negative breast cancer were treated as experimental group.Of the remaining 301 breast cancer patients,69 cases were randomly assigned to the control group.Immunohistochemistry was used to detect the expression of ROR1 in triple negative breast cancer and other subtypes of breast cancer.And analyzed the relationship between ROR1 expression and clinicopathological features and prognosis in triple negative breast cancer patients.Results: 1.The positive rate of ROR1 in breast cancer was 73.9%(102/138).The positive rate of triple negative breast cancer was 91.3%(63/69),the high expression rate was 57.9%(40/69),and the low expression rate was 33.3%(23/69).In other subtypes of breast cancer,the positive rate was 56.6%(39/69),the high expression rate was 15.9%(11/69),and the low expression rate was 40.6%(28/69).ROR1 expression in triple negative breast cancer was significantly higher than that in other subtypes of breast cancer,and the difference was statistically significant(P < 0.001).2.ROR1 expression was associated with lymph node metastasis,histological grading,TNM staging,and ki-67 expression in patients with triple negative breast cancer,the difference was statistically significant(P< 0.05).But no significant correlation wasfound with patients' age,site,menstrual status,age of menstruation,and tumor size(P >0.05).3.In the survival analysis with triple negative breast cancer,the DFS and OS were65.2%and 85.5%,respectively.The DFS and OS of ROR1 high expression group were significantly lower than that of ROR1 non-high expression group,and the difference was statistically significant(P < 0.05).Univariate analysis of survival analysis showed that,DFS was correlated with lymph node metastasis,histological grade,TNM stage,ki-67 expression and ROR1 high expression(P < 0.05);OS was correlated with lymph node metastasis,histological grade,TNM stage and ROR1 high expression(P < 0.05).Multivariate COX proportional hazard regression analysis showed that ROR1 expression,histological grade and TNM stage were independent influencing factors of patients' DFS(P < 0.05),but ROR1 expression was not significantly correlated with OS(P>0.05).Conclusion:1.ROR1 has a higher overall expression rate in breast cancer and is significantly higher in triple negative breast cancer than other subtypes of breast cancer.2.Lymph node metastasis,histological grading,TNM staging,ki-67 expression were correlated with ROR1 expression in triple negative breast cancer patients.3.ROR1 expression,histological grading and TNM staging were independent influencing factors of patients' DFS.
