Clinical And Basic Study On The Prevention And Treatment Of Postoperative Stone Recurrence And Cholelithiasis With Dahuang Lingxian Capsule

Objective: To observe the effect of Dahuang Lingxian Capsule on the contraction function of gallbladder and the recurrence rate of gallstone in patients with gallstone after cholelithiasis and to explore the mechanism of its prevention and treatment of gallstone in mice.Methods:(1)clinical study: from January 2018 to December 2019,60 patients after cholelithiasis were selected from hepatobiliary surgery of the First Affiliated Hospital of Guangxi University of traditional Chinese medicine.They were divided into control group(30 cases)of Xiaoyan Lidan tablet and control group(30 cases)of Dahuang Lingxian capsule.On the first day after operation,the patients in the control group were given 6 Xiaoyan Lidan tablets three times a day,and the patients in the treatment group were given6 Dahuang Lingxian capsules three times a day for six months.The systolic function of gallbladder,thickness of gallbladder wall,recurrence rate of calculus,blood routine(WBC,CRP),urine routine and stool routine,and renal function of heart and liver(alt,AST,etc.)were observed after 6 months of treatment.(2)Basic experiment: 60 C57 BL / 6 mice were divided into normal group(N Group),model group(M group),ursodeoxycholic acid capsule group(U group)and Dahuang Lingxian capsule group(D group)according to random number table.In addition to the normal group,other groups of mice were fed with high-fat,high cholesterol(15%fat,1% cholesterol,5% cholic acid)diet.According to the weight equivalent drug dose conversion of mice and adults,group u was given 130 mg / kg · D UDCA powder solution by gavage,group D was given 13 g / kg · D rhubarb Lingxian capsule solution by gavage.Eight weeks later,the gall bladder of mice was dissected to observe the stone formation rate.The liver tissues were taken for PCR and Western blotting to detect the gene and protein expression differences of liver inflammatory factors IL-6,IL-8 and TNF-?.Conclusion: Dahuang Lingxian capsule can improve the gallbladder contraction function and the thickness of gallbladder wall,reduce the recurrence of calculus,and has no side effect.At the same time,Dahuang Lingxian capsule can reduce the expression of IL-6,IL-8,TNF-? inflammatory factors in the liver of mice,reduce liver damage,reduce the production of pathological bile,promote bile excretion,and thus play a role in the prevention and treatment of stones.
