Objective : In recent years,the incidence rate of thyroid carcinoma has increased dramatically with the detection rate of thyroid benign and malignant tumors increasing.Among them,papillary thyroid carcinoma(PTC)takes the first place in the total proportion of thyroid carcinoma.The majority of PTC is well differentiated malignant tumor,which manifests as mild clinical behavior and has a good prognosis,it still brings physical pain,mental pressure and even over treatment to many patients.At the same time,some PTC patients with early cervical lymph node metastasis or extragland?Lar invasion lead to poor prognosis,and the risk of tumor recurrence increases after treatment,which affects the quality of life of patients.PTC is mainly treated by surgery at present,but accurate preoperative diagnosis,individualized assessment and postoperative monitoring are facing severe challenges due to lack of specific molec?Lar biomarkers.Long non-coding RNA(lncRNA)is a kind of new molec?Lar biomarker,which has been confirmed to be involved in the development of PTC,and it may be used as a potential molec?Lar biomarker to predict the indicators of PTC.In order to provide help for the diagnosis and treatment level of PTC,the expression situation and clinical significance of lncRNA ITGB-1 were preliminarily explored,and its possible mechanism was analyzed in this paper.Methods : Clinical part : 34 cases of PTC specimens and correspondingadjacent normal tissues were collected after surgery from September 2018 to January 2020 in the Affiliated Hospital of Southwestern Medical University,and the expression of lncRNA ITGB-1 and its relation with clinic pathology were analyzed by Quantitative Real-time PCR.Cell part: Thyroid papillary carcinoma cell TPC-1 and normal thyroid cell Nthy-ori3-1 were c?Ltured,and RT-qPCR was used to detect the expression level of lncRNA ITGB-1 in TPC-1 cells and Nthy-ori3-1 cells.SiRNA was used to down-reg?Late the expression of lncRNA ITGB-1.The effect of lncRNA ITGB-1 on the invasion of TPC-1 cells was studied by transwell experiment.The effect of lncRNA ITGB-1 on the migration of TPC-1 cells was studied by cell scratch test.CCK8 experiment was used to investigate the effect of lncRNA ITGB-1 on the proliferation of TPC-1 cells.It was predicted that the down-reg?Lation of lncRNA ITGB-1 might lead to the biological mechanism of PTC by searching online literatures and biological websites.Res?Lts:The expression of lncRNA ITGB-1 in papillary thyroid carcinoma tissues was higher than that in corresponding adjacent normal tissues(P<0.05),the expression of PTC tissues with lymph node metastasis and membrane or extragland?Lar invasion was significantly higher than that had no above characteristics(P < 0.001,P < 0.01).The expression of lncRNA ITGB-1 in TPC-1 cells was significantly higher than that in Nthy-ori3-1cells(P < 0.05).The expression of lncRNA ITGB-1 in TPC-1 cells co?Ld be inhibited by siRNA(P < 0.01).Down-reg?Lating the expression of lncRNA ITGB-1 co?Ld inhibit the invasion of TPC-1 cells.Down-reg?Lating the expression of lncRNAITGB-1 co?Ld inhibit the migration of TPC-1 cells.Down-reg?Lating the expression of lncRNA ITGB-1 cloud inhibit the proliferation of TPC-1 cells.It was predicted that down-reg?Lating the expression of lncRNA ITGB-1 co?Ld inhibit the expression of snail factor,which co?Ld inhibit epithelial mesenchymal transformation(EMT)to delay the development of thyroid papillary carcinoma.Conclusion : The expression of lncRNA ITGB-1 in papillary thyroid carcinoma tissues was higher than that in corresponding adjacent normal tissues,and it was more significant in papillary thyroid carcinoma with lymph node metastasis and membrane or extragland?Lar invasion.The expression of lncRNA ITGB-1 in TPC-1 cells was higher than that in Nthy-ori3-1 cells.Down-reg?Lating the expression of lncRNA ITGB-1 co?Ld inhibit the invasion,migration and proliferation of TPC-1 cells,which suggested that lncRNA ITGB-1 was related to the development of PTC.Down-reg?Lating the expression of lncRNA ITGB-1 might inhibit the expression of snail factor,which co?Ld inhibit epithelial mesenchymal transformation(EMT)to delay the development of thyroid papillary carcinoma.In a word,lncRNA ITGB-1 might be regarded as a potential molec?Lar biomarker to guide the diagnosis and treatment of PTC after further study.
