Objectives To investigate the expression of Lnc RNA TCONS00003757 in papillary thyroid carcinoma and explore the effect of Lnc RNA TCONS00003757 on proliferation,migration and invasion in TPC-1 cells.Methods 60 cases of papillary thyroid carcinoma specimens and paired normal tissue were collected after surgery from October 2017 to March 2018 in the Tangshan Workers Hospital.RT-PCR analysis was used to examine the expression of Lnc RNA TCONS00003757 in PTC tissues and cell lines,and the relationship between Lnc RNA TCONS00003757 expression and the patients' clinicopathological data was analyzed.Furthermore,the effect of Lnc RNA TCONS00003757 and ST3GAL5 silencing on cell proliferation,migration and invasion were assessed in PTC in vitro by MTT,scratch test and transwell assay.Finally,protein expression of ST3GAL5 was detected by Western blot.Results LncRNA TCONS00003757 was overexpressed in PTC tissues and TPC-1 cells?P < 0.05?.Lnc RNA TCONS00003757 expression was correlated to capsule invasion and lymph node metastasis?P<0.05?.Silencing the expression of Lnc RNA TCONS00003757and ST3GAL5 inhibited the proliferation,migration and invasion of thyroid cephalic cancer cells,and the reducing of ST3GAL5 expression was related to Lnc RNA TCONS00003757silencing?P<0.05?.Conclusions Lnc RNA TCONS00003757 expression in papillary thyroid carcinoma was significantly increased and correlated with the occurrence and development of thyroid gland.Silencing Lnc RNA TCONS00003757 can significantly inhibit the invasion and migration of thyroid papillary cancer cells,and ST3GAL5 protein may be the target of its action.Figure7;Table1;Reference 124...
