Objective:To observe the effects of electroacupuncture(EA)stimulation on gastric movement and autophagy of interstitial cells of Cajal(ICC)in DGP model rats,so as to investigate its mechanism of EA in the treatment of DGPMethods:Healthy SD rats were randomly divided into blank group and model group.The DGP models were induced by single intraperitoneal injection of streptozotocin and by feeding the rats with high-sugar and high-fat diets.Those rats which established successfully were again randomly divided into model group,EA group and metoclopramide group.The EA group was performed to"Liangmen" "Zusanli" and "Sanyingjiao".The metoclopramide group were received 1.7%concentration of metocloppramide(1mL/100g).The blank group and model group were received 0.9%sodium chloride solution(1mL/100g)Record blood glucose of all rats for every week.After the treatment,the gastrointestinal propulsive rate were measured by intragastric phenol red,the content of IGF-1 and EGF in gastric antrum were detected by ELISA,the expression of LC3 in gastric antrum was detected by weston blot,and the autophagy level was observed by transmission electron microscopeResults:1.Compared with the blank group,the blood glucose in the model group was significantly increased and the gastrointestinal propulsive rate were significantly decreased(P<0.01);Compared with the model group,the blood glucose in the EA group was significantly reduced and the gastrointestinal propulsive rate were significantly increased(P<0.05 or P<0.01),the gastrointestinal propulsion rate in the metoclopramide group were increased significantly(P<0.01)and the blood glucose did not change significantly(P>0.05);Blood glucose and gastrointestinal propulsive rate were not significantly different between electroacupuncture group and gastric resuscitation group(P>0.05).2.Compared with the blank group,the contents of IGF-1,LC3-?/LC3-? in the gastric antrum tissue of the model group increased significantly,and the EGF content decreased significantly(P<0.05 or P<0.01);Compared with the model group,the contents of IGF-1,LC3-?/LC3-? in the gastric antrum tissue of the EA and metoclopramide group decreased significantly,and the EGF content increased significantly(P<0.05 or P<0.01);There was no significant difference in the contents of IGF-1,EGF,LC3-?/LC3-I in the gastric antrum tissue of the EA group compared with the metoclopramide group(P>0.05).3.The ICC structure of gastric antrum in the blank group was intact and no obvious autophagic structure was found;In the model group,there was an autophagosome and ICC was severely injured;In the EA group,there were 2 autolysosomes and the ICC injury was better than model group;In the metoclopramide group,there were 2 autophagic vesicles and the ICC injury was better than model groupConclusion:1.EA at“Liangmen”“Zusanli”and“Sanyingjiao”could improve symptoms of DGP rats,decrease blood glucose and improve gastrointestinal motility.2.DGP rats develop gastrointestinal dysmotility mainly because the fusion of autophagosome and lysosome is blocked,and then autophagy flux impaired.3.EA may improve the gastrointestinal motility of DGP rats by promoting the fusion of autophagosomes and lysosomes to ease the impaired autophagy flux.The possible mechanism is that EA can regulate the contents of LC3-?/LC3-??IGF-1 and EGF in gastric antrum.
