Effect Of Shiweixiaopi Decoction On Gastric Antral Interstitial Cells Of Cajal And SCF Protein Expression In Diabetic Gastroparesis Rats

Objective: To investigate the possible role of Interstitial Cells of Cajal and SCF protein expression in the pathogenesis of diabetic gastroparesis and clarify the possible mechanism of Shiweixiaopi Decoction for the treatment of diabetic gastroparesis.Methods: 50 healthy male Wistar rats were randomly divided into 4groups:normal comparison group(NG),DGP model group(MG),Shiweixiaopi Decoction group(ZG)and Mosapride citrate group(XG)for experimental research.Diabetic gastroparesis model in rats were established by a single intraperitoneal injection of streptozotocin and high-energy diet and irregular feed.The general condition,body weight and the blood glucose of all rats were detected and gastric residual rate was measured by gastrogavage with semisolid paste.To observe the pathological changes of gastric tissue by microscope.The expression and distribution characteristics of c-kit antibody positive ICC was detected by immunohistochemical method in gastric antrum tissue of rats in each group.And the average optical density(A)value of c-kit was detected by the biological electrophoresis image.And taken the Western blot to detect gastric tissue SCF protein expression level.Results:1 The general stateCompared with NG,the rats appeared polydipsia,polyuria and more food,after modeling,and continuing.But gradually activity decreased,eating less,hair dryness.After drug intervention,the general condition of rats,compared with MG,all had different levels of improvements,especially in ZG.2 The change of body weight in ratsCompared with NG,the body weight in MG,ZG and XG all reducedobviously,statistically significant difference(P<0.05);compared with MG,the body weight of rats in ZG and XG increased,statistically significant difference(P<0.05);compared with XG,the weight gain was more obvious in ZG,statistically significant difference(P<0.05).3 The change of blood glucose levelThe blood glucose level of rats in MG,ZG,XG was highter than that in NG,statistically significant difference(P<0.05);compared with MG,the blood glucose level of ZG was decreased,statistically significant difference(P<0.05);the blood glucose level of XG was lower than that in MG,no statistically significant difference(P>0.05)?4 The gastric residual rate in ratsThe gastric residual rate in NG was the lowest when compared with the other 3 groups,statistically significant difference(P<0.05);compared with MG,the gastric residual rate in ZG and XG was obviously decreased,statistically significant difference(P<0.05);compared with XG,the gastric residual rate of ZG was significantly decreased,statistically significant difference(P<0.05).5 HE staining results of gastric smooth muscle cellsThe gastric antrum mucosa structure of NG was integrated and distinct,rich of blood vessels,the gland arranged neatly,the smooth muscle cells in spindle shape,distributed a lot of interstitial cells;compared with NG,the gastric antrum mucosa of MG was structural damaged,the number of blood vessels was decreased,glands disorganized,smooth muscle cells became smaller,distribution of interstitial cells decreased significantly;in ZG and XG,the mucosa was complete,glands arrangement was neat,blood vessels were richer,most smooth muscle cells in spindle shape,a certain amount of interstitial cells.6 The expression of positive c-kitOn the mean optical density comparison of positive c-kit,compared with the other 3 groups,NG was the highest,statistically significant difference(P<0.05);the mean optical density in ZG and XG was highter than that in MG,statistically significant difference(P<0.05);the mean optical density and expression level were higher in ZG than in XG,statistically significant difference(P <0.05),but failed to reach the expression level of the ICC in NG.7 The expression level of SCF proteinCompared with NG,the expression level of was obviously decreased in MG,ZG,XG,statistically significant difference(P<0.05);after the intervention of drugs,the SCF protein expression level was higher in ZG,XG than in MG,statistically significant difference(P <0.05);compared with XG,the SCF protein expression level of ZG was increased,statistically significant difference(P <0.05).Conclusion:1 Shiweixiaopi Decoction,not only can improve the DGP rats general situation,but also increase the mean optical density of positive c-kit and the expression level of SCF protein,is an effective method for the treatment of diabetic gastroparesis;2 Shiweixiaopi Decoction may rebuild the pathway by the upregulation of the ICC and SCF expression,so as to enhance contraction force of gastric smooth muscle cells and promote gastric emptying.
