ObjectiveThis research was aimed to investigate the association between the four single nucleotide polymorphisms of CDH13 gene and blood glucose level of the elderly.MethodsThrough cross-sectional study,a health survey was conducted on the elderly in a county in Hechi City,Guangxi Zhuang Autonomous Region.A case-control study was conducted in 603 patients with high fasting plasma glucose and 3504 healthy controls.The DNA in the blood sample of the study subject was extracted by Aidlab kits.Four SNPs of CDH13 gene(rs6565105,rs72807847,rs58409508,rs8051657)were genotyped using imLDRTM multiplex SNP kits.Statistical analysis was performed using SPSS22.0 statistical software.The measurement data was analyzed by t test,analysis of variance and other statistical methods.Chi square test was used for categorical data.The Hardy-Weinberg genetic equilibrium was tested by Plink software.The genetic environment interaction analysis was performed using GMDRv9 software.All of above tests were performed using two-sided tests,test level ?=0.05.Results1.General demographic characteristics:A total of 4017 elderly people were included in the study.The average age of the population was 70.02,and the number of males was less than that of females.Zhuang was the main ethnic group with a total of 3,521 people,accounting for 85.73%of the total population.The smoking rate in the general population was 15.36%,and the drinking rate was 22.55%,and both rates were higher for men than for women.2.Influencing factors of fasting plasma glucose(FPG):The study was conducted in 603 patients with high fasting plasma glucose(hyperglycaemia group)and 3504 healthy controls.Gender,ethnicity,age,height,weight,body mass index(BMI),waist circumference(WC),FPG,high-density lipoprotein cholesterol(HDL-c),triglycerides(TG),systolic blood pressure(SBP),diastolic blood pressure(DBP)and alcohol consumption were statistically different between the two groups(P<0.05).Male(OR=0.691,95%CI=0.553?0.864),age(OR=1.029,95%CI=1.017?1.042),WC(abdominal obesity)(OR=2.054,95%CI=1.558?2.707),drinking(OR=1.424,95%CI=1.117?1.815),and TG(OR=1.669,95%CI=1.248?2.233)five factors were risk factors for fasting plasma glucose.3.Hardy-Weinberg equilibrium test:The four SNPs of CDH13 gene were consistent with Hardy-Weinberg equilibrium in the control group(P>0.05).4.Allele frequency and genotype distribution:In the general population,the allele and genotype distribution of rs58409508 and rs8051657 of CDH13 gene were statistically different between the hyperglycaemia group and the control group(P<0.05),while there was no statistical significance in rs6565105 and rs7280784(P>0.05).In the female population,the alleles and genotypes of the rs6565105 and rs58409508 of the CDH13 gene were statistically different between two groups(P<0.05).And there was no significant difference in the allele and genotype distribution of each SNPs in males between two groups OP>0.05).5.Correlation between CDH13 SNPs and FPG in different gene models:In the dominant gene model of the rs6565657,the risk of impaired fasting glucose(IFG)in the CT+CC genotype carrying population was 0.785(0.659?0.935)times that of the TT genotype,which was consistent in female(ORF=0.75095%CI:0.580?0.969).In the co-dominant and dominant gene models of rs58409508,taking the GG genotype as a reference,carrying GA genotype,AA genotype,GA+AA genotype increased the risk of impaired fasting glucose(IFG)(ORGA=1.420,ORAA=12.723,ORGA+AA=1.482).In the recessive model,taking the GG+GA genotype as a reference,carrying the AA genotype increased the risk of IFG,OR=12.501,95%CI:1.127?138.723.6.The effect of the interaction between CDH13 gene and environmental factors on FPG:In the general population,rs58409508 polymorphism,WC and alcohol consumption were the optimal interaction models.In the female population,WC and rs58409508 polymorphism became the optimal model.In the general population,the combination of GG genotype carrying at the rs58409508,non-drinking,and normal waist circumference is a protective factor for IFG.In the female population,the combination of high or normal WC with AA genotype carrying at the rs58409508 and high waist circumference with GA and GG genotypes at the rs58409508 are risk factors for IFG.7.Linkage disequilibrium and haplotype analysis:The linkage disequilibrium between the four sites of rs6565105,rs72807847,rs58409508 and rs8051657 was lower.There were significant differences in the distribution of GAGT and GAAT between the hyperglycaemia group and the control group(P<0.05).Compared to the control group,the GAGT frequency was lower and the GAAT frequency was higher in the hyperglycemic group.8.Analysis of different SNPs and lipid metabolism in serum:In the hyperglycemia group,genetic polymorphisms at the rs6565105 and rs8051657 sites were associated with HDL-c levels in serum.In the control group,the rs6565105 locus gene polymorphism was associated with TG levels in serum.Conclusion1.The proportion of IFG in the elderly in a county of Guangxi was low.Age,male,waist circumference(abdominal obesity)and alcohol consumption are risk factors for IFG in the elderly.2.The gene polymorphisms of rs58409508 and rs8051657 of CDH13 gene might be associated with increased fasting blood glucose in the elderly in a certain county of Guangxi.And the polymorphisms of rs6565105 and rs8051657 might be associated with decreased HDL-c levels in people with IFG.3.The polymorphism of CDH13 gene rs58409508 had an interaction with WC and drinking,which might be related to the increase of fasting blood glucose in the elderly in a certain county of Guangxi.
