Liangxue Tongyu Decoction Can Regulate Gastrointestinal Hormones And Metabolites In Rats With Intracerebral Hemorrhage To Exert An Anti-Inflammatory Effect

Objective:To study the mechanism of regulating gastrointestinal hormones and metabolites in rats with intracerebral hemorrhage.Methods:Intracerebral hemorrhage rat model was prepared by autologous tail-artery blood injection.SD rats were randomly divided into sham operation group,model group and cooling blood stasis group,and the relevant indicators were observed at three time points of 24h,72h and 120h after administrationThe behavior score of rats with cerebral hemorrhage was measured by Longa grade 5 score method,and the moisture content of brain tissue was measured by dry and wet weight method.The expressions of TNF-a and il-lb in brain tissues were detected by ELISA,and the expressions of NF-KB in brain tissues were detected by immunohistochemistry.The expressions of gastrin(GAS),motilin(MTL)and cholecystokinin(CCK)in brain tissues were detected by ELISA.The expressions of nitric oxide(NO),carbon monoxide(CO),hydrogen sulfide(H2S)and d-lactic acid in serum of rats were detected by ELISA.Results:1.Effects on brain damage:(1)behavioral score:the medication of liangxuetongyu prescription group was 72h and 120h,and the score of Longa grade 5 was lower than that of the model group(P<0.05).(2)water content of brain tissue:72h and 120h after administration,the cooling blood circulation stasis group was significantly lower than the model group(P<0.01),and there was no difference between the sham operation group and the cooling blood circulation stasis group(P>0.05).2.Anti-inflammatory effect:(1)the content of TNF-a(815.94,646.12,397.92 pg/ml)was significantly lower in the model group(1240.98,1037.55,867.29 pg/ml)and higher in the sham group(288.92,263.61 pg/ml)(P<0.01)at 24h and 72h than that in the sham group(260.53 pg/ml)(P>0.05).(2)for 24h,72h,and 120h,the content of il-1b in the liangxuetongyu group(863.83,623.65,517.53 pg/ml)was significantly lower than that in the model group(1279.76,1213.69,973.91 pg/ml)(P<0.05,P<0.01),and that in the sham group(307.91,294.21 pg/ml)for 24h and 72h was significantly higher than that in the sham group(325.53 pg/ml)(P>0.05).(3)for 24h,72h and 120h,the content of nf-kB P65(7.15,5.53 and 4.52 pg/ml)in the liangxuetongyu group was significantly lower than that in the model group(9.45,9.12 and 7.73 pg/ml)(P<0.05,P<0.01),and there was no difference between the sham operation group and the sham operation group(4.37 pg/ml)(P>0.05).3.Results of the determination of gastrointestinal hormones in brain tissues:(1)gastrin(2878.39 pg/g)in the treatment group and the model group(2615.64pg/g)showed no difference(P>0.05).Medication for 72h and 120h(3000.75,2695.08 pg/g)were significantly higher than that of the model group(2349.95,1989.59 pg/g)(P<0.01),and there was no difference between the model group(1989.59 pg/g)and the sham operation group(1763.19 pg/g)(P>0.05).(2)after 24h of administration,there was no difference in motilin(3607.59 pg/g)between the model group and the model group(3195.03 pg/g)(P>0.05),and both 72h and 120h of administration(3523.61,3115.69 pg/g)were higher than that of the model group(2626.96,2309.61 pg/g)(P<0.05,P<0.01).(3)there was no difference between the model group(1909.81 pg/g)and the cooling blood circulation group(2081.89 pg/g)for 24h(P>0.05),and there was no difference between the model group(1759.69,1557.41 pg/g)and the model group(1391.78 pg/g)for 72h and 120h(2069.08,1794.98 pg/g)(P<0.05,P<0.01).4.Serum metabolite determination results:(1)the carbon monoxide(CO)concentration(28.46,25.11,22.97 umol/L)in the liangxuetongyu group was lower than that in the model group(33.65,35.09,32.59 umol/L)(P<0.05,P<0.01).(2)for 24h,72h and 120h,the concentration of nitric oxide(NO)in the liangxuetongyu group(46.95,42.11 and 30.32 umol/L)was higher than that in the model group(29.90,26.32 and 19.36 umol/L)(P<0.01),and that in the model group(19.36 umol/L)was lower than that in the sham operation group(24.40 umol/L)(P<0.05).(3)24 h,72 h,120 h,cool through blood stasis group of hydrogen sulfide(H2S)concentration(7.96,8.81,9.81 nmol/L)were higher than in model group(5.91,6.25,5.57 nmol/L)(P<0.01),the model group were lower than that of control group(8.99,9.82,10.20 nmol/L)(P<0.05,P<0.01),and 120 h cool blood stasis group(9.81 nmol/L)and control group(10.20 nmol/L)no difference(P>0.05).(4)there was no difference in d-lactic acid concentration(37.73 umol/L)between the treatment group and the model group(36.00 umol/L)(P>0.05).After treatment for 72h and 120h,the concentration of the cooling blood circulation stasis group(33.71,25.07 umol/L)was lower than that of the model group(39.42,37.49 umol/L)(P<0.01),and there was no difference between the cooling blood circulation stasis group(25.07 umol/L)and the sham operation group(26.28 umol/L)(P>0.05)..Conclusion:Cooling blood circulation formula can reduce the water content of brain tissue in rats with cerebral hemorrhage,reduce the cerebral edema,and improve the nerve function damage.Liangxue tongyu prescription can inhibit the activity of nf-kb inflammatory signaling pathway,reduce the expression levels of TNF-a,il-1b and other inflammatory factors,and thus reduce the damaging inflammatory response.lt can increase the secretion of GAS,MTL,cck-8 and other brain intestinal peptides in the brain tissue,promote intestinal perikinetic function,reduce the expression of inflammatory factors secreted by macrophages stimulated by the accumulation of endotoxin in the blood,and play an anti-inflammatory role.Maintain the low concentration of CO expression,the synthesis and release of the neurotoxic metabolite d-lactic acid,promote the synthesis and release of NO and H2S,and inhibit the expression of inflammatory factors.To sum up,the prescription of "liangxuetong yu" plays an anti-inflammatory role through the enteric-cerebral axis neuroendocrine and direct communication pathways.
