Objective:To study the effect of total flavonoids of Dracocephalum Moldavica L.(DML)on chronic mountain sickness(CMS)in rats by ~1H-NMR metabolomics.Methods:After successfully build the model of CMS,the experimental rats were randomly divided into 6 groups(n=8),control group(CG,saline 0.4 mL/100 g/d),model group(MG,saline 0.4 mL/100 g/d),the nifedipine group(NE,nifedipine tablets 2.7 MG/kg/d),total flavonoids of DML high-dose,medium-dose and low-dose group(DML.H,DML.M,DML.L,were respectively given 400 mg/kg/d,total flavonoids of DML 200 mg/kg/d,100mg/kg/d of total flavonoids of DML).The serum of rats in each group was measured for NMR metabolomics analysis.Serum levels of apolipoprotein A1(Apo-A1)and apolipoprotein E(Apo-E)were measured.The brain histopathological changes of rats in each group were observed under low power and high power lens,respectively.Results:The serum test results showed that the total flavonoids of DML can significantly increase the serum levels of apolipoprotein A1 and apolipoprotein E in rats with chronic mountain sickness.The results of serum metabolomic analysis showed that total flavonoids of DML can improve amino acid metabolism disorder,energy metabolism disorder and lipid metabolism disorder in rats with chronic mountain sickness.The results of pathological examination showed that total flavonoids of DML can improve the pathological changes in brain tissues of rats with chronic mountain sickness,such as meningeal vasodilation,hyperemia,brain parenchymal edema,inflammatory cell infiltration,enlarged perivascular space and enlarged pyramidal cells.Conclusion:Total flavonoids of Dracocephalum Moldavica L has potential therapeutic effect on chronic altitude sickness,and the possible mechanism is improve metabolic disorder and reduce oxidative damage.
