Research background:Colorectal cancer is a common malignant tumor with high morbidity and mortality,which seriously affects human life.Surgical treatment is the preferred method,but the growth of colorectal cancer is relatively slow.From the occurrence of the tumor to the occurrence of clinical symptoms,the tumor has experienced a long period of growth and development.Therefore,chemotherapy plays an important role in the treatment of colorectal cancer.However,there are differences in drug efficacy between different individuals,and many patients are not sensitive to chemotherapy drugs.Traditional Chinese medicine holds that "blood stasis is necessary for long-term illness".The clinical manifestations of tumor patients are mostly stasis.Promoting blood circulation and removing blood stasis is an important method to treat stasis.Most traditional Chinese medicines and compound prescriptions for promoting blood circulation and removing blood stasis have certain inhibitory effects on tumorsObjective:This topic uses the method of network pharmacology to obtain the network of"compound formula-effective components of drugs-target genes-diseases" for treating colorectal cancer with Qinghua Anticancer Decoction,to explore the nature of biological pathway for treating colorectal cancer with Qinghua Anticancer Decoction,to predict its mechanism of action,and to provide help for clinical research and treatment.Methods:The pharmacokinetic ADME parameters were used for screening,the effective components of Qinghua Anticancer Decoction were screened out in the database,the prediction of traditional Chinese medicine components and targets was completed through TCMSP,and target gene annotation was carried out.At the same time,disease targets are screened out to form gene intersection with them,PPI network is constructed by using network database platforms(TTD,OMIM,STRING),core genes of the network are screened out,GO and KEGG enrichment analysis is carried out by using Biocondutor software,and the possible action pathway of Qinghua anticancer decoction for adjuvant treatment of colorectal cancer is obtained Result:Taking bioavailability(OB%)?30%and drug-like property(DL)?0.18 as parameters,76 effective components of Qinghua Anticancer Decoction were screened out,and 199 core targets of colorectal cancer were obtained by PPI network architecture through STRING.The GO and KEGG enrichment analysis of predicted targets was carried out by Bioconduclor software,involving 173 signal pathways,of which chromosome instability pathway and microsatellite instability pathway are the two main types.Qinghua anticancer decoction may inhibit cell hyperproliferation and stimulate mitochondria to induce apoptosis to take effect on tumor through receptor pathway,which indicates that Qinghua anticancer decoction plays a multi-target,multi-angle and multi-level role in the treatment of colorectal cancer and provides direction for subsequent clinical research.
