| Colorectal cancer(CRC)is one of the most common gastrointestinal tumors in general surgery.Due to the lack of effective molecular diagnosis and therapeutic targets,once the disease is diagnosed,patients are basically in the middle and advanced stages.Currently,the main treatment method is surgical treatments,and the prognosis is not ideal.Long non-coding RNA(lnc RNA)belongs to a class of non-coding RNAs with the length of about 220 nt.It has been shown that lnc RNA plays a significant role in the occurrence and development of colorectal cancer.However,the function of lnc RNA AC007392.4 in colorectal cancer is unclear.Therefore,this study systematically explores its biological function in colorectal cancer for the first time,and preliminary explores the potential molecular mechanism of its biological function in colorectal cancer.Objective: Colorectal cancer is one of the common malignant tumors in China,and exploring the molecular mechanism of its occurrence and development is the main direction of colorectal cancer research.Methods: Long non-coding RNA AC007392.4 expression in colorectal tissue was analyzed by the Cancer Genome Atlas(TCGA),and then the expression of AC007392.4in colorectal cancer was verified by clinical samples;Finding the protein-coding genes located downstream of AC007392.4 through the calculation of Pearson correlation,and performed enrichment analysis on these protein-coding genes in biological processes and biological pathways;AC007392.4 vector and si RNA were designed and synthesized,and transfected into SW480 by liposome transfection method.Cell functional experiments were performed on AC007392.4 in colorectal cancer cell.Finally,the effect of AC007392.4 on the mRNA of key molecules in the downstream pathway was detected by RT-qPCR.Results:1)The TCGA database shows that AC007392.4 is up-regulated in colorectal cancer and is related to the pathological stages of colorectal cancer patients.Clinical samples also show that AC007392.4 is highly expressed in colorectal cancer;2)Correlation analysis shows that a total of 2354 protein-coding genes were highly correlated with AC007393.4 expression,and these protein-coding genes are significantly enriched in DNA damage repair and cell cycle-related pathways;3)CCK-8 experiments and apoptosis experiments show that AC007392.4 promotes the proliferation of colorectal cancer cells while inhibiting their apoptosis;4)Cell cycle related experiments show that knocking down AC007392.4 can block G2 /M phase of cell cycle;5)RT-qPCR results showed that after knocking down AC007392.4,the expression level of cell cycle molecule CDK1 mRNA was significantly reduced(P <0.05).Conclusion: AC007392.4 increased expression in colorectal cancer,promotes the proliferation and inhibits apoptosis of colorectal cancer cell,and may participate in the development of colorectal cancer by regulating the cell cycle.This study provides a potential theoretical basis for the clinical diagnosis and molecular targeted therapy of colorectal cancer. |
