Objective:This subject takes nutritional obese rats as the research object,and uses molecular biological detection to analyze the effect of "tonify the kidney and replenish qi,fortify the spleen and harmonize stomach" acupuncture to intervene with the protein expression of adipose tissue,so as to reveal the correlation between acupuncture intervention and adipose tissue protein expression in obese rats.Methods: According to the random principle,10 male SD rats were selected as a blank control group and fed with ordinary feed,the remaining rats were fed with a high-fat diet to establish a nutritional obese rat model,and 20 rats were selected from the successful model rats.As acupuncture treatment group and obesity model group,each group had 10 animals.Then use "tonify the kidney and replenish qi,fortify the spleen and harmonize stomach" acupuncture to intervene in the acupuncture treatment group.acupuncture once a day for 4 consecutive weeks.During the treatment period,the rats in the blank control group and the obese model group were also restrained and fixed every day,but there was no acupuncture intervention.After 4 weeks of intervention,the blood was collected from the rat aorta to detect serum albumin and blood lipid levels,and the wet weight of adipose tissue around the intestine,kidney,and epididymis was measured,and the protein expression of adipose tissue around the intestine was detected by immunobltting.Results:(1)Changes in the survival status,body weight,wet weight ofvisceral adipose tissue,and blood lipids of rats in each group: After intervention,the general conditions of the acupuncture treatment group,such as mental state,activity,hair color,diet,and stool,were significantly improved compared with the obesity model group.Before treatment,all indexes in the obesity model group were significantly higher than those in the blank control group(P<0.05),and the differences were statistically significant.After the intervention treatment,the values of all indexes in the acupuncture group were significantly lower than those before the intervention(P<0.05),the indexes of the rats in the acupuncture group were significantly lower than those in the obesity model group(P<0.05).(2)Changes of protein expression in the pericytes of a rat in each group:Compared with the blank control group,the expressions of TGFBR1,TGFBR2,SMAD3,ITGB1 and GFAP in serum albumin and pericyta in the obese model group were significantly up-regulated,Compared with the obesity model group,the expression levels of TGFBR1,TGFBR2,SMAD3,ITGB1 and GFAP in serum albumin and peri-intestinal adipose tissue were significantly decreased in the acupuncture treatment group.Conclusion:(1)Acupuncture with "tonify the kidney and replenish qi,fortify the spleen and harmonize stomach" as a method can obviously improve the living condition of obese rats,reduce the body weight,the wet weight of adipose tissue around the internal organs and the level of blood lipid.(2)In the obesity model group,serum albumin,TGFBR,SMAD3,ITGB1 and GFAP are highly expressed and positively correlated with each other,suggesting that these proteins participate in the development of obesity,and there may be albumin/TGFBR/SMAD/ITGB1/GFAP signaling pathway in the body.(3)In the acupuncture treatment group,serum albumin,TGFBR,SMAD3,ITGB1 and GFAP were all low expressed and positively correlated with each other,indicating that acupuncture treatment may reverse the albumin/TGFBR/SMAD/ITGB1/GFAP signal Upregulation of the pathway Inhibits obese rats.
