Backgroud and Objective :Postoperative ileus is a common complication after abdominal surgery.Its mechanism has not been fully determined,and there is still a lack of effective drugs and means for prevention and treatment,Houpu Sanwu Tang As a traditional Chinese medicine,has been found that it may have a certain degree of improvement for postoperative ileus in recent years.This research selected some potential targets and mechanisms for the effect of Houpu Sanwu Decoction on postoperative ileus through network pharmacology.Observe and exploer the effect of Houpu Sanwu Tang on postoperative ileus and the expression of these potential targets by POI rat model.To provide some evidence and clues for Houpu Sanwu Tang improve postoperative ileus.Methods:The potential gene targets related to Houpu Sanwu Tang and postoperative ileus were screened by using network pharmacology.60 SD Male rat aged 8-10 months were randomly divided into 5 groups: sham group,model group,high-dose group,medium dose group,low-dose group.Sham group make a incision in the abdominal wall not enter the abdominal cavity,other groups constructed POI rat model through manipulated the small bowel.At the 6,12,24,36 h after operative,high-dose group,medium dose group,low-dose group intragastric treatment of different dose Houpu Sanwu Tang.Sham group and model group treated with normal saline of equal volume.At the 48 h after operative,selected 6 rats from each group given 1ml carbon ink for intragastric treatment,30 minutes later,take out the small bowel,measure the length of carbon ink dyeing and the total length of the small bowel,calculate the small bowel propulsion rate.For the remaining 6 rats in each group,take material from their terminal small bowel,Observe their morphologica changes by microscope and The expression level of potential gene targets,which were screened by network pharmacology,were analyzed by using western blot,ELISA experimental techniques.Results:1.Three possible gene targets IL-6,AKT1 and VEGFA,which were associated with Houpu Sanwu Tang and postoperative ileus,were obtained by network pharmacology.2.The postoperative ileus model rat experimental results showed that: the small bowel propulsion rate and the edema of high-dose group,medium dose group and low-dose group were significantly better than the model group.3.HE staining experiment results showed that: small bowel villus state of high-dose group,medium dose group and low-dose group were significantly better than the control group.4.The results of the western blot experiment showed that: compared with the model group,the small intestine tissue AKT1 and VEGFA protein expression level showed a significant downward adjustment in the high-dose group and medium dose group.5.ELISA experimental results showed that: compared with the control group,the small bowel tissue expression level of IL-6 protein showed a significant reduction in the high-dose group and medium dose group.Conclusion:Houpu Sanwu Tang can significantly improve the postoperative ileus in rats,and its mechanism may be related to the down-regulation of the expression of AKT1,VEGFA and IL-6..
