Objectives:To study the possible mechanism of curcumin enhancing cisplatin in inhibiting the invasion and metastasis of human lung cancer A549 cells,so as to provide a new target for the treatment of lung cancer.Methods:Culture human lung cancer A549 cells in vitro and randomly divide them into five groups:Curcumin group,cisplatin group,curcumin + cisplatin group,miR-218 inhibitor group and negative control group,Process each group as required.The expression of miR-218 in A549 cells was detected by RT-PCR,and the expression of each Slug,ZEB2 and E-cadherin was detected by Western blotting.Results:1.The relative quantity of miR-218 in curcumin group,cisplatin group,curcumin + cisplatin group,negative control group and miR-218 inhibitor group were:1.753 ± 0.033,2.021 ± 0.115,3.0640 ± 0.149,1.0430± 0.058 and 0.2690 ± 0.016,Among them,curcumin group,cisplatin group,curcumin + cisplatin group and miR-218 inhibitor group were comparedwith the negative control group respectively,curcumin group,cisplatin group,curcumin + cisplatin group and miR-218 inhibitor group were compared with each other,curcumin + cisplatin group was compared with curcumin group,curcumin + cisplatin group was compared with cisplatin group,curcumin + cisplatin group was compared with miR-218 inhibitor group,Comparison between group differences are statistically significant(P < 0.05).2.The IOD of protein in curcumin group,cisplatin group,curcumin + cisplatin group,negative control group and miR-218 inhibitor group,Slug was:38.64 ± 3.18,28.79 ± 2.88,16.29 ± 1.87,52.88 ± 4.65,66.64 ± 3.87,respectively;the expression of ZEB2 was:34.92 ± 4.22,26.47 ± 1.78,15.35 ± 2.03,49.48 ± 2.08,56.78 ± 0.55,respectively;whether Slug or ZEB2,the four different experimental groups compared with the negative control group,among the four groups,curcumin +cisplatin group and curcumin group,curcumin + cisplatin group and cisplatin group,curcumin + cisplatin group and miR-218 inhibitor group,Comparison between group differences are statistically significant(P < 0.05).3.The IOD of E-cadherin protein in curcumin group,cisplatin group,curcumin + cisplatin group,negative control group and miR-218 inhibitor group was 15.38 ± 0.92,21.33 ± 1.22,40.78 ± 1.65,10.38 ± 1.20,7.79 ± 0.95,respectively.the four different experimental groups comparedwith the negative control group,among the four groups,curcumin +cisplatin group and curcumin group,curcumin + cisplatin group and cisplatin group,curcumin + cisplatin group and miR-218 inhibitor group,Comparison between group differences are statistically significant(P <0.05).Conclusions:1.Curcumin and cisplatin both enhanced the expression of miR-218 in A549 cells,and the combination effect was better.2.Curcumin and cisplatin both down-regulated the expression of Slug and ZEB2 in A549 cells,up-regulated the expression of E-cadherin in A549 cells,and the combination effect was better.3.The mechanism of curcumin enhancing cisplatin in inhibiting the invasion and metastasis of A549 cells may be closely related to the enhancement of miR-218 expression,down-regulation the expression of Slug and ZEB2,and up-regulation the expression of E-cadherin.
