Objective:At present,it has been found that AIM2(absent in melanoma 2)gene and intestinal flora participate in the development of colorectal cancer.However,it is not clear whether there is a correlation or even syn ergy between the two in the pathogenesis of colorectal cancer.In this stud y,16SrRNA sequencing of preoperative fecal samples from patients with colorectal cancer was performed to identify the structure and relative abu ndance of intestinal flora,and the expression of AIM2 in tumor tissues was used as a group basis to analyze the expression of AIM2 gene and composition of intestinal flora.Is there a correlation?Methods:A total of 59 preoperative stools of inpatients diagnosed with colorectal cancer from September 2018 to January 2019 were collected fr om Sichuan Provincial People's Hospital,and 59 cases of surgically confi rmed colorectal cancer were diagnosed by pathology1.Obtain stools of colorectal cancer patients during hospitalization throug h sterile cups,requiring stools treated for 4 days or more or stools with out bowel treatment.The collection process must be aseptic,and refrigerat ed after low temperature treatment2.Obtain cancer tissue specimens from the Department of Pathology,Sic huan Provincial People's Hospital.The specimens are clear specimens cont aining cancer tissues and subjected to MMR gene immunohistochemical e xperiments.AIM2 gene-specific immunohistochemical staining(EnVision method)was performed to determine the results with the help of patholo gist The classification and comparative analysis of the AIM2 staining res ults of each tissue and its relationship with the clinicopathological data o f patients3.Immunohistochemical methods involved:The study uses an enzyme-labe led antibody method,specifically horseradish peroxidase(HPR)and a sec ondary antibody to prepare a conjugate.When the primary antibody and t he substrate to be detected After antigen-specific binding,a secondary ant ibody is added to form an enzyme-polymeric compound-second antibody complex,and a specific color reaction occurs through diaminobenzidine4.Intestinal flora sequencing method:by obtaining the stool of the patient before surgery,extracting genomic DNA after processing,and performin g PCR amplification on the bacterial 16SrRNA gene sequence V4 hyperv ariable region,and high-throughput sequencing analysis on lluminaMiSeq platform The proportion and type of flora composition5.Analyze the collected clinical and pathological data of patients with aim2 staining score spss17.0 statistical software for statistical analysis,and use the R software to statistically process and analyze the Intestinal flora data.Results And Conclusion:The study included 59 patients with colorectal cancer,including 36 males(61.02%)and 23 females(38.98%).The avera ge age of the patients was 63.84±13.21 yearsThe pathological specimens of 59 colorectal cancer patients were ana lyzed by immunohistochemistry,and the expression of aim2 in tumor tiss ues was found.The patients with tnm stage I/IV were lower than those i n stage ?/?.It is suggested that patients with low expression of aim2 in c olorectal cancer tissues have a late clinical pathological stage and a wors e prognosisBy 16S RNA sequencing and analysis of preoperative intestinal flora in patients with colorectal cancer,it was found that the composition of intestinal flora in patients with colorectal cancer is different from that of normal human intestinal flora.The colony diversity and relative abundanc e of the intestinal flora of AIM2 high expression group and AIM2 low e xpression group were similar,suggesting that the expression of AIM2 wa s not related to the overall composition of the intestinal floraSpecies analysis between groups showed that at the gate level,the a verage abundance of the thick-walled bacteria in the AIM2 high expressi on group was higher than that of the AIM2 low expression group(591%vs 46.9%,p=0.0284);at the genus level,AIM2 The average abundan ce of Blautia and Faecalibacteriium in the high expression group was hig her than that in the low expression group of AIM2,while the average a bundance of Peptostreptococcus,Eisenbergiella,Tyzzerella and Megasphaer a was lower than that in the low expression group of AIM2.Both were statistically significant.This suggests that some bacterial abundance change s may be related to the expression of the AIM2 gene.
