Objective: Absent in melanoma 2(AIM2)is a kind of double stranded DNA receptor found in recent years,which can induce inflammatory response and immune response by the formation of inflammatory complexes,and is closely related to the occurrence and development of malignant tumor.AIM2 has a certain degree of abnormal expression in the malignant tumor tissue,and the expression of AIM2 may be related to the clinicopathological features and prognosis of patients with malignant tumor,but the way and mechanism of its involvement in tumorigenesis is not clear,and there is no relevant data in our country.This study intends to investigate the expression of AIM2 gene in tumor tissues of patients with colorectal cancer.and explore the relationship between AIM2 and clinicopathological features of colorectal cancer.Methods: From 2016 January to 2016 June,patients with resectable primary colorectal cancer were involved and their clinical materials recorded.After tumor was resected,the tumor tissues and adjacent normal tissues were taken for AIM2 immunohistochemical staining,the expression of AIM2 in colorectal cancer tissues and adjacent normal tissues were observed under the microscope.complete pathological data of patients were recorded,to analyze the relevance between the expression of AIM2 and the pathological data of colorectal cancer patients.This experiment use enzyme labeled polymermethod,horseradish peroxidase is first labeled on dextran polymer,then form a Envision two resistance with the second antibody link.AIM2 specific antibodies combined with tissue antigens.Let Envision two antibody combine with the first antibody of the antigen-antibody complex,the expression of AIM2 in the tissues was analyzed by the Envision two antibody enzyme in the color reaction.Results: The clinical and pathological data of 100 patients with colorectal cancer were analyzed,there were 57 male patients and 43 female,with an average age of 62.88±10.56 years old.Immuohistochemical staining showed that the positive expression of AIM2 protein was mainly in the cytoplasm of intestinal epithelial cells;1.Tumor tissues and adjacent normal tissues in 100 patients with colorectal cancer,the number of AIM2 positive expression cases were 42(42%)and85(85%).Indicating that the expression of AIM2 in colorectal cancer tissues is significantly lower than that in adjacent normal tissues,the difference is statistically significant(?2=39.888;P<0.05);2.Compared with positive expression,the patients with negative expression of AIM2 in the tumor tissues,with worse T staging(depth of tumor invasion)?N staging(lymph node metastasis)and tumor stage,and more prone to mismatch repair protein negative expression(dMMR)(?2=4.408,4.995,6.565,3.923;P<0.05).But there was no significant correlation between expression of AIM2 in colorectal cancer tissues and gender?age?tumor location?tumor size?tumor differentiation degree and tumor pathological type.(?2=0.375,0.642,0.270,0.037,1.002,1.376;P>0.05).Conclusion:1.The AIM2 protein is mainly located in the cytoplasm of colorectal epithelial cells;2.The expression of AIM2 in the colorectal cancer tissues is significantly lower than that in adjacent normal tissues,indicating that AIM2 may be a potential tumor suppressor;the low expression or lack of AIM2 may be an important factor contributing to the occurrence and development of colorectal cancer;3.Colorectal cancer patients with low expression of AIM2 in tumor tissues have worse pathological stage,which may lead to worse prognosis;4.Colorectal cancer patients with low expression of AIM2 in tumor tissue,the incidence of dMMR is more higher,suggesting that the decrease or deletion of AIM2 expression may be the consequence of gene mutation caused by MMR deletion.
