| Objective: By observing the effects of Yinchen Wuling Powder on the expression levels of sterol regulatory element binding protein-1c(SREBP-1c)and nuclear transcription factor-кB(NF-κB)in non-alcoholic fatty liver disease rats,the effects of Yinchen Wuling Powder on NAFLD were explored The mechanism of action provides laboratory basis for its clinical application.Method: 70 male SD rats were randomly divided into a blank control group(15 rats)and an experimental group(55 rats).The blank control group was given normal feed,and the rats in the experimental group were fed with modified high-fat feed to make a model of non-alcoholic fatty liver disease,which was fed for 24 weeks.After successful modeling,the rats in the experimental group were randomly divided into 5 groups: non-alcoholic fatty liver group,Yinchen Wulingsan 1.28 group,Yinchen Wulingsan 0.64 group,Yinchenhao Decoction group,simvastatin control group,each group 10 pcs.Rats in each group were administrated by gavage and given corresponding drug treatment.After 6 weeks,the liver mass,body mass,liver index,low density lipoprotein cholesterol(LDL-c),triglyceride were measured in each group.Ester(Triglyceride,TG),total cholesterol(TC),Aspartate Aminotransferase(AST),Alanine Aminotransferase(ALT)levels,using HE staining to observe liver pathological morphology changes;fluorescence Quantitative polymerase chain reaction(RT-PCR)was used to detect the expression of SREBP-1c m RNA and NF-κB m RNA in liver tissues.Result: Compared with the blank control group,the expressions of TG,LDL-c,AST,ALT,body mass,liver index,liver mass,TC,SREBP-1c and NF-κB in the non-alcoholic fatty liver group were increased(P <0.01).Compared with the non-alcoholic fatty liver group,the expression of rats in the simvastatin control group,Yinchenhao decoction group,Yinchen Wulingsan 1.28 group and Yinchen Wulingsan 0.64 group were all reduced(P <0.01).Compared with the simvastatin control group,all the indexes of Yinchen Wulingsan 1.28 group and Yinchenhaotang group were significantly lower(P <0.01),and Yinchen Wulingsan 0.64 group LDL-C,TC,TG,SREBP-1c and NF-κB Both were significantly reduced(P <0.01),liver quality and liver index were decreased(P <0.01,P <0.05),body weight difference was not statistically significant(P> 0.05),ALT and AST were decreased(P <0.05,P <0.01).Compared with Yinchenhao Decoction group,Yinchen Wulingsan 1.28 group ALT,AST,liver quality,liver index and body mass,SREBP-1C,NF-κB were significantly reduced(P <0.01),there was no difference in LDL-C,TC and TG Statistical significance(P> 0.05),Yinchen Wuling powder 0.64 group ALT,AST increased(P <0.05,P <0.01),LDL-C,TC,TG,SREBP-1C,NF-κB,liver quality and body weight Significantly increased(P <0.01),and there was no statistically significant difference in liver index(P> 0.05).Conclusion: Yinchen Wuling Powder can improve the lipid metabolism of hepatocytes,slow down the degree of steatosis,inhibit inflammation,oxidative stress,and apoptosis of hepatocytes by regulating the expression levels of SREBP-1c and NF-κB in rats,thereby protecting liver tissues and treating Non-alcoholic fatty liver disease.Yinchen Wuling Powder 1.28 group is superior to Yinchen Wuling Powder 0.64 group in preventing and treating non-alcoholic fatty liver disease,and the treatment effect of Yinchen Wuling Powder is better than Yinchenhao Decoction at the same drug concentration and dose. |
