| Objective: This study focused on the dampness,heat,and blood stasis pathogenesis of diabetes combined with non-alcoholic fatty liver disease,and explored the treatment of Yinchenhao Decoction,the main prescription of Clearing heat,removing dampness and removing stasis on diabetes combined with non-alcoholic fatty liver disease.The intervention effect of Zhang Zhongjing’s Clearing away heat,removing dampness and removing stasis method and the relevant provisions of Yinchenhao Decoction are sorted out and summarized,and Zhang Zhongjing’s law of Clearing heat,removing dampness and removing stasis method is discussed in depth with Yinchenhao Decoction as the main prescription.Under the guidance of Zhang Zhongjing’s Clearing heat,removing dampness and removing stasis method,MKR mice as the test subjects,and Yinchenhao Decoction was used as a representative prescription to conduct animal experiments.By observing the effect of Yinchenhao Decoction on the pathological changes and inflammatory response of MKR mice’s liver to explore the guiding role of Zhang Zhongjing’s method of clearing and removing blood stasis in the treatment of diabetes with non-alcoholic fatty liver disease.Method:(1)Theoretical research part.Discussing the etiology of diabetes and non-alcoholic fatty liver disease in Chinese medicine and the related theories of pathogenesis.Summarizing the specific content of Zhongjing’s method of Clearing heat,removing dampness and removing stasis,expounding the specific application of the representative Yinchenhao Tang,and analyzing Clearing heat,removing dampness and removing stasis the modern application value of Clearing heat,removing dampness and removing stasis method for diabetes combined with non-alcoholic fatty liver disease.(2)Experimental research part.Fifty 8-week-old MKR mice were selected,10 were blank controls,and 40 were fed with high-fat diet for 8weeks to establish a model.After modeling,randomly divided into 4 groups:model group,Yinchenhaotang original prescription group,Yinchenhaotang group and positive control group(metformin + simvastatin).Mice were sacrificed after 8 weeks administration,and measured by electrochemical method glucose tolerance,serum inflammatory IL-6,TNF-α measured by ELISA method,blood lipid and liver function changes measured by blood biochemical analyzer,liver tissue morphology was observed by transmission electron microscope and HE staining,liver tissue was detected by Western blot Changes in TLR4,MyD88,and NF-κB protein expression.Results: Theoretical results show that the main pathogenesis of diabetes with non-alcoholic fatty liver is the accumulation of damp heat and blood stasis,and the main pathological factors are dampness,heat,and blood stasis.The Yinchenhao Decoction created by Zhongjing has the effect of clearing heat,removing dampness and removing blood stasis.It has a solid theoretical basis for the treatment of dampness,heat,and stasis-based diabetes combined with non-alcoholic fatty liver disease.The results of animal experiments showed that compared with the model group,the glucose tolerance of the drug group was improved(P<0.01),the serum IL-6and TNF-α levels were reduced,and the liver inflammatory cells The expression of TLR4,MyD88 and NF-κB protein was down-regulated in the Yinchenhaotang group and the Yinchenhaotang original prescription group(P<0.05).Conclusion:1.The main pathological factors of diabetes with non-alcoholic fatty liver are dampness,heat,and blood stasis,and the pathogenesis of TCM is damp-heat and stasis resistance.Yinchenhao Decoction can intervene with the method of Clearing heat,removing dampness and removing stasis as a representative effect.2.Yinchenhao Decoction can reduce serum inflammatory factors and blood lipid levels in MKR mice,and down-regulate the protein expression of liver TLR4,MyD88,and NF-κB.3.Yinchenhao Decoction can improve the pathological state of type 2diabetes combined with non-alcoholic fatty liver disease by reducing liver inflammation and improving liver function. |
