Objective: To explore the etiology and pathogenesis of perimeno pausal syndrome,the current status of TCM treatment and its co rrelation with apoptosis through theoretical research;establis h an ovariectomized animal model.Serum sex hormone and blood l ipid content,uterine cell apoptosis and detection of apoptosis factors Bcl-2,Bax,Cyto-C,to explore the possible mechanism of treatment of perimenopausal syndrome with Bushenhuatan presc ription,provide more theory for clinical treatment And experim ental basis.Methods: Fifty healthy 6-month-old SPF female rats were randoml y divided into a blank control group,a sham operation group,a model group,a kidney and phlegm and blood stasis group,and a Livial group.There were 5 groups of 10 rats in each group.An ovary-removing animal model was established and a vaginal smea r examination was performed.Those without keratosis had bilate ral ovaries completely removed,that is,the model was successf ul.The blank control group,sham operation group and model gro up were given 0.9% sodium chloride solution,the kidney and phl egm and blood stasis group was given 940mg/ml of liquid solutio n,and the Livial group was given 0.023mg/ml Livial suspension,1ml/100 g.Twenty-four hours after the last administration,the rats underwent intraperitoneal anesthesia.After satisfactory anesthesia,they collected blood from the heart and collected a s much blood as possible in the anticoagulation tube.Above the rat pubis,a "Y" type tissue,that is,rat uterine tissue,was immediately removed,fixed with 4% paraformaldehyde,and froze n at 4 ° C until use.Calculate the weight of ovariectomized r ats,observe the uterine morphology of the rats under the micro scope by HE staining method,detect the content of E2,FSH,LH and TC,TG,HDL-C,LDL-C in the serum of rats by ELISA method,detect by IHC The protein expression of Bcl-2,Bax and Cyto-C i n rat uterine cells was analyzed quantitatively,and the apopto sis rate of rat uterine cells was detected by TUNEL method.Results: 1.Compared with the sham operation group,the body we ight of the model group increased significantly,with a signifi cant difference(P<0.05).Compared with the model group,the bo dy weight of the rats in the Bushen Huatan Quyu group and the L iweiai group decreased significantly,the difference was statis tically significant(P<0.05).2.Compared with the sham operation group,the model group's uterus was significantly reduced,the endometrial atrophy was thinned,the glands were mostly low columnar,the number was v ery small,the muscle layer was thinned,and the serous layer w as atrophy and not smooth.Compared with the model group,the v olume of the uterus in the kidney-enriching phlegm group was si gnificantly increased.The endometrium in the hyperplasia p eriod had regular muscle layers and smooth serous membrane laye rs.3.Compared with the sham operation group,the E2 level of the model group was significantly reduced,and the FSH and LH l evels were significantly increased,with a significant differen ce(P<0.01).Compared with the model group,the serum E2 level was increased in the kidney-tonifying phlegm group and the livi al group,and the serum FSH and LH levels in the two groups dec reased,with a significant difference(P<0.01 or P<0.05).4.Compared with the sham operation group,the content of T C,TG,LDL-C in the model group was significantly increased,an d the level of serum HDL-C was significantly reduced,the diffe rence was statistically significant(P<0.01 or P<0.05).Compare d with the model group,serum levels of TC,TG and LDL-C were s ignificantly reduced in the kidney-tonifying phlegm group and t he livial group,while the serum HDL-C levels of the two groups increased,with a significant difference(P<0.01 or P< 0.05).5.Compared with the sham operation group,the Bcl-2 protei n expression of the model group was significantly reduced,and the protein expression of Bax and Cyto-C was significantly incr eased,with a significant difference(P<0.01).Compared with th e model group,the Bcl-2 protein expression of the kidney-enric hing phlegm group and the Livial group was significantly increa sed,and the protein expression of Bax and Cyto-C of the two gr oups decreased,with a significant difference(P<0.01 or P<0.05).6.Compared with the sham operation group,the apoptosis ra te of the model group was significantly increased,and there wa s a significant difference(P<0.05).Compared with the model gr oup,the apoptotic rate of the Bushen Huatan Quyu group and Liw eiai group decreased significantly,and the difference was stat istically significant(P<0.05).Conclusion: Bushen Huatan Quyu Recipe can reduce the weight and improve uterine morphology of ovariectomized rats,regulate se x hormone levels and lipid metabolism,up-regulate the expressi on level of anti-apoptotic proteins Bcl-2,and down-regulate pr o-apoptotic proteins Bax and Cyto-C The level of expression red uces the rate of apoptosis of uterine cells,and its mechanism of action is likely to be through the regulation of Bcl-2/Bax/C yto-C signaling pathway protein-mediated cell and mitochondrial apoptosis.
