BackgroundEndometrial cancer is one of the three major malignant tumors of reproductive tract,and the most common histological type is adenocarcinoma from endometrial glands.In recent years,the incidence of endometrial cancer has been increasing worldwide,and with the rapid development of the global economy,the number of obesity and diabetes has increased significantly,resulting in an increasing proportion of young endometrial cancer patients.A considerable number of patients could not receive surgery,radiotherapy and chemotherapy because of their multiple complications or their own fertility requirements,resulting in the loss of treatment opportunities,poor quality of life and short survival time.Therefore,clinicians have been trying to.find new treatment that can not only only achieve efficacy but also improve the life quality of patients.Myoblasts transplantation therapy(MTT)is a new type of gene therapy in the world.A large number of studies have shown that myoblasts transplantation can repair the damaged gene of degenerated muscle fiber,supply the living cells,and strengthen the systolic and diastolic function of myocardial cells,which has a certain effect on the treatment of muscular malnutrition and myocardial diseases.In animal experiments,it also shows that myoblasts have a certain therapeutic effect on diabetes.In recent years,MTT has also been shown to have a therapeutic effect on animal prostate cancer models,which indicated that MTT might serve as a new therapeutic method for cancer.ObjectiveThe purpose of this study is to understand the influence of myoblasts transplantation therapy on endometrial cancer and to explore new therapeutic method for endometrial cancer by comparing such effects as changes in weight and tumor volume,morphological changes,and immunohistochemistry of injection of myoblasts,cisplatin and normal saline on endometrial cancer model.Methods(1)Thirty nude mice models with endometrial cancer were established and randomly divided into three groups,and then injected with normal saline,cisplatin and myoblasts,respectively.(2)After injection,the weight of nude mice and the length and short diameter of tumor were measured every other day for 4 weeks.(3)Four weeks later,HE staining and immunohistochemistry were performed to observe the tumor tissue morphology and detect the expression of tumor related proteins.Results(1)Before the injection,the body weight and tumor volume of nude mice in normal saline group,cisplatin group and myoblast group were(17.13±0.35 g,36.34±0.11 mm3),(17.12±0.34 g,36.32±0.12 mm3),(17.03±0.29 g,36.27±0.09 mm3),respectively,and there was no significant difference in body weight and tumor volume between the three groups(P>0.05).Four weeks later,the body weight and tumor volume of normal saline group,cisplatin group and myoblasts group were(16.58±0.19 g,96.51±0.26 mm3),(17.35±0.19 g,45.18±0.25 mm3),(17.97±0.08 g,21.21±0.14 mm3),respectively.The body weight of the nude mice in the normal saline group decreased and the tumor body increased.The body weight of the mice in the cisplatin group increased slightly,but the tumor growth was not significant.The body weight of nude mice in myoblast group increased greatly and the tumor body decreased.And there was significant difference among the three groups(P<0.05).(2)HE staining showed that there were a large number of endometrial cancer cells with obvious heteromorphism in the tumor tissue of normal saline group,with rapid cell proliferation and a small amount of necrosis in the center of the tumor tissue.In cisplatin group,there were some necrosis in the tumor tissue.And in myoblastic group,there were a large number of tumor tissue necrosis,cell necrosis and nuclear lysis could be seen under the microscope.(3)The results of immunohistochemistry showed that the positive rate of HER2 was 50%in the saline group,30%in the cisplatin group and 10%in the myoblast group,and there was no significant difference in HER2 positive rate among the three groups(P=0.149).Conclusion1.Compared with the normal saline group,the weight of the myoblasts group increased,the tumor volume decreased,and the positive rate of HER2 was low,which suggested that myoblasts were an effective method for the treatment of endometrial cancer.2.Compared with cisplatin in the treatment of endometrial cancer,myoblasts were better than cisplatin.
