| Objective:The goal of the present experimental study is to investigate effect on adult rabbit autologous skeletal myoblasts injection into the thyroarytenoid(TA) muscle after recurrent laryngeal nerve (RLN) injury and scar vocal cordMethods:Adult big white rabbit,s cleidomastoidal mascle myoblasts was cultivated,segregated and expand . The left side big white the rabbit neck was make a horizontal nick, long about 4 cm, One by one in order cut open a skin, subcutaneous tissue, separate ahead of the neck muscle group, and find out recurrent laryngeal nerve in the ditch of windpipe oesophagus at the thyroid gland bottom, at the 2-4 tracheal ring level cut off recurrent laryngeal nerve 1 m, the near heart recurrent laryngeal nerve was sew in subcutaneously and sew up nick after taking out left side cleidomastoidal mascle (provide cleidomastoidal mascle myoblasts) .it was to expose glottis under rising epiglottis with the laryngoscope and row a scar at the left side vocal cord surface wihh the hooked needle, the depth arrived vocal cord muscle. The established animal model about recurrent laryngeal nerve (RLN) injury and scar vocal cord.20 big white rabbit random were divided into 2 groups, each 10, 20 big white the rabbit all were cut off recurrent laryngeal nerve (RLN) and hurted the left side vocal cord . A group was injection skeletal myoblasts in the left thyroarytenoid (injection skeletal myoblasts group),the right side was normal compare; B group was injection Sodium Chloride in the left thyroarytenoid in the left thyroarytenoid (injection Sodium Chloride group), the right side was normal compare. To examine A group and B group thyroarytenoid muscle fiber diameter and vocal cord volume after 12 weeks.Results:Didn't show difference(p>0.05) from itself double muscle fiber diameter and vocal cord volume of injection autologous skeletal myoblasts and did show difference (p<0.05) from double muscle fiber diameter and vocal cord volume of injection Sodium Chloride after 12 weeks.Conclusion:injection autologous skeletal myoblasts in thyroarytenoid muscle made myoblasts become obvious increase and live around thyroarytenoid muscle fiber after 12 weeks from immunohistochemistry under Microscope and thyroarytenoid muscle fiber diameter become thickening and vocal cord volume become biger and restore close not glottis of recurrent laryngeal nerve (RLN) injury and scar vocal cord . |
