The Study Of Therapy Of Fructus Xanthium For Allergic Rhinitic

ObjectiveAmong the 30 commonly used Chinese tradition medicine prescriptions for the treatment of rhinitis,sinusitis and allergic rhinitis,23 of them contain Fructus Xanthii.It has the effects of dispersing wind cold,nasal pass and eliminating wind damp.In this research,we combined the animal pharmacology experiments in vivo and cell pharmacology experiments in vitro to observe the effects of fructus Xanthii for AR.For example,observe the effect of Fructus Xanthii water extract on the AR model guinea pigs,observe the effects of Fructus Xanthii containing serum on ?-hexosaminidase and histamine secreted by cell allergy model,the effects on the release of interlenkin-4,interlenkin-17 and interferon-? from cell inflammation models.Methods1.The guinea pigs animal model was sensitized with a suspension of ovalbumin and aluminum hydroxide.The experiments were divided into: normal group,model group,loratadine group,Yupingfeng group,and high-,middle-,low-dose fructus Xanthii water extract groups(4,2 and 1 g/m L,respectively).After the model was established successfully,the scores of nasal behavioral symptoms were observed under a light microscope.Elisa was used to measure the serum leves of immunoglobulin E,interleukin-4,and interferon-?.2.Ig E sensitized and bovine serum albumin stimulated RBL-2H3 cell to establish a cell allergy model.The experiments were divided into normal group(normal basophils),normal serum group(10% normal guinea pig serum),model group,normal serum control group(0.5 ?g/m L Ig E and 10% normal guinea pig serum),2h group,4h group,6h group,12 h group,24 h group.MTT method was used to detect the proliferation of RBL-2H3 cell and the effect of Fructus Xanthii containing serum on the viability of RBL-2H3 cell.The release of ?-hexosaminidase from antigen-stimulated RBL-2H3 cell was measured.And the histamine release was measured by Elisa method.3.Induce the cell model of RAW264.7 cell as an inflammatory reaction by using lipopolysaccharide.The experiments were divided into normal group(normal macrophages),normal serum group(10% normal guinea pig serum),model group,normal serum group(20?g/m L LPS and 10% guinea pig serum),2h group,4h group,6h group,12 h group,24 h group.MTT method was used to detect the proliferation of RAW264.7 cell and the effect of Fructus Xanthii containing serum on the viability of RAW264.7 cell.Elisa method was used to detect the contents of IL-4,IL-17 and IFN-?.Results1.Compared with that of the normal group,the behavioral scores of guinea pig in model group increased,and there was significant inflammatory cell infiltration and glandular expansion in the nasal mucosa.Compared with the model group,the high-,middle-and low-dose Fructus Xanthii water extract reduced serum Ig E and IL-4,and increased IFN-? in a dose-dependent manner.2.MTT results showed that there was no significant difference in cell proliferation between the 2h Frutus Xanthii containing serum in the 10%-50% concentration range and the normal group serum.Compared with the control group,the degranulation rate of RBL-2H3 cell in the model group was increased significantly,and the release levels of histamine was increased significantly,too.The ?-hexosaminidase release rate and histamine release were significantly reduced in RBL-2H3 cell treated with Fructus Xanthii containing serum.3.MTT results showed that there was no significant difference in cell proliferation between the 2h Frutus Xanthii containing serum in the 10%-50% concentration range and the normal group serum.Compared with the normal group,the model group after LPS intervention increased the high expression of IL-4,IL-17,and reduced the release of IFN-?.Compared with the model group,the serum of Fructus Xanthii containing different groups reduced the expression of IL-4,IL-17,and increased the release of IFN-?.ConclusionThe results of animal experiments showed that Fructus Xanthii could remarkably alleviate the AR symptoms,the morphological changes of nasal mucosa,and reduced serum Ig E,IL-4,and increased IFN-?.The results of cell experiments showed that Fructus Xathii containing serum can significantly reduce ?-hexosaminidase and histamine levels,can significantly reduce the levels of IL-4,IL-17,and increase the level of IFN-?.
