Objective:To investigate the effect of the bark extract of Sambucus sibirica Nakai on bone metabolism and growth factor in fracture model rats,and to explore its mechanism.Methods: 108 3-month-old male SD rats were randomly divided into 9 groups,12 rats in each group: sham operation group,model control group,positive drug Dida pill group,alc ohol extract high,medium and low dose groups,water extract high,medium and low dose groups.Blood samples and femur samples were taken after 2,4 and 6 weeks of drug interv ention,respectively,to observe the effects of two extracts of Sambucus sibirica Nakai on the indexes of serum bone metabolism,he staining and the expression of BMP-2 and VEG F in bone tissue of fracture end.Results: compared with the blank control group,the level of ALP,OPG,ACP,BGP and PICP in the model group increased significantly(P < 0.05);c ompared with the B group,the level of ALP,ACP,BGP and PICP in the d 1,d 2,e 1,e 2 a nd c group increased significantly(P < 0.01,P < 0.05)The level of ALP,ACP,BGP and P ICP decreased with the process of fracture healing,while the content of OPG increased wi th the process of fracture healing,the difference was statistically significant(P < 0.05).Li ght microscopy showed that the number of new bone trabeculae increased in group d1,e1 and c,forming homogeneous bone trabeculae,and osteoblasts and osteoclasts were seen.I n group d2,d3,e2,e3 and b,a small amount of new bone trabeculae and less osteoid were found.The positive expression of BMP-2 and VEGF in bone tissue of fracture end was ma inly found in the cytoplasm of bone cells in mesenchymal cells,fibroblasts,osteoblasts,ch ondrocytes and osteoid,and the staining was deeper in high dose group;the expression of BMP-2: compared with group b,drug intervention 14 The difference was statistically signi ficant(P < 0.01,P < 0.05)in d1 and e1 groups at d and 42 d,but significantly higher in d1,e1 and c groups at 28 d after drug intervention(P < 0.01,P < 0.05).Compared with group c,the difference was statistically significant(14 days,42 d,P < 0.05;28d,P < 0.01).VEG F expression: after 14 days of drug intervention,compared with group b,group d1,d2,e1,c significantly increased,the difference was statistically significant(P < 0.01 in group d1,e1;P < 0.05 in group c,d 2).At 28 d,compared with group B,group d1,d2,e1,e2,cwere significantly higher(P < 0.01;P < 0.05),compared with group c,group D 1,E 1 were hig her than the model group,and the difference of d1 was statistically significant(P < 0.05);at 42 d,compared with group b,group d1,e1,c were statistically significant(P < 0.05).Co nclusion: Sambucus sibirica Nakai bark extract can increase the content of bone metabolism index in serum,make the situation of bone metabolism better,promote the exp ression of BMP-2 and VEGF,and promote the fracture healing,and the effect of d1,e1 an d c group is stronger than that of other groups,among which the effect of d1 group is stron ger than that of e1 and C group.The mechanism of action may be to promote the proliferat ion of osteoblasts and accelerate the process of fracture healing by promoting the expressi on of BMP-2 and VEGF in bone tissue.
