Objective:To study the anti-atherosclerotic efficacy and mechanism of Aifeitimeng Tang by using a high-fat diet to reproduce atherosclerotic rabbit model.Methods:(1)Acute toxicity test:120 mice were randomly divided into 5 control groups(normal saline)and Efitimone Decoction,a total of 6groups,20 in each group,half male and half male;The state of mice was observed continuously for 14 days.(2)A high-fat diet method was used to establish an AS rabbit model,and at the same time,rabbits were fed with Efitimone decoction for prevention.(3)Observe the pathological changes of rabbit aorta by HE is staining.(4)Detection of total cholesterol(TC),cholesterol lipid(TG),low-density lipoprotein(LDL-C),high-density lipoprotein(HDL-C)in serum of AS rabbits,and the use of a 721 tube photometer to measure super oxidation in serum Contents of dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),nitric oxide(NO),and malondialdehyde(MDA).(5)The effects of aortic proliferating cell nuclear antigen(PCNA)and platelet-derived growth factor(PDGF-A)were measured by immunohistochemical method and Western blot method was used to detect the effect of Aifeitimeng Tang on atherosclerotic rabbits.Effects of Arterial ?B(NF-?B)and PCNA Protein Expression.Results:(1)Gross anatomy examination after acute poisoning experiment:gastrointestinal congestion or flatulence were found in the dissection observation of dead animals;gross dissection was performed on animals of each group after 14 days of observation,and no abnormal tissues and organs were seen by naked eyes.The acute toxicity LD50 of Aifeitimeng Tang mice was 10.7g/kg,and the average confidence interval was 9.4 to 12.0g/kg.(2)Under the light microscope,the thickness of arterial vascular wall in the model group can be seen to be different,the endometrium is obviously thickened,foam cells are proliferated,granulation tissue formation and lymphocyte infiltration are seen around it,and the median smooth muscle is locally hyperplastic.There are atherosclerotic lesions.Serum TC,TG and LDL-C in rabbits fed a high-fat diet were significantly increased,and the AS rabbit model was successfully established.The Aifeitimeng Tang group significantly reduced serum TG,TC,and LDL-C and other indicators,increased SOD,GSH-PX,CAT,and NO,and reduced MDA content(P<0.05-0.01).The results of immunohistochemistry showed that the expressions of PDGF and PCNA in rabbit aorta in the model group were significantly increased.Compared with the model group,the expression of PCNA in each group of Aifeitimon Decoction was significantly reduced,and there was a certain dose-effect relationship(among them Low dose group P<0.05,medium and high dose group P<0.01);Aifeitimeng Tang low and high dose group PDGF positive rate significantly decreased(P<0.05);Western Blot results showed that high dose group PCNA expression was significant Decrease(P<0.05).Conclusion:The acute toxicity LD50 of Aifeitimeng tang mice is 10.7g/kg,which is 331 times the daily dosage per adult body weight of adults.Aifeitimeng Tang can reduce the content of TC,TG,LDL-C in the serum of AS rabbits,increase the effects of SOD,GSH-PX,CAT and NO,and reduce the content of MDA;the mechanism is by reducing the PCNA and PDGF of the blood vessel wall The content of-A decreases the expression of PCNA protein in the vascular wall of rabbits,and reduces the expression of NF-?B in the blood vessels,thereby inhibiting rabbit proliferating cell nuclear antigen and platelet-derived growth factor and preventing the occurrence of AS.
