Objective:To explore the effect and mechanism of acyclovir(ACV)on the migration and apoptosis of human eye TENON capsule fibroblasts(HTFs)in vitro.Methods:HTFs were divided into ACV-treated groups(final concentrations:0.45,1.125,2.25,3.375,4.5 mmol·L-1)and blank control groups.CCK8 was used to detect the effect of ACV on the proliferation rate of HTFs under different concentration gradients.The scratch test was used to detect the effect of ACV at different concentrations on the migration of HTFs cells.Flow cytometry was used to detect the effects of ACV on HTFs apoptosis and cell cycle.Results:Compared with the blank control group,the proliferation rate of HTFs in the ACV-treated group(final concentrations were 0.45,1.125,2.25,3.375,4.5 mmol·L-1,respectively)was significantly reduced(P<0.05).With the increase,the inhibition rate of HTFs gradually increased.In the ACV-treated group(final concentration of 4.5mmol·L-1),the apoptosis rate increased significantly(P=0.0005),the peak value of the cell cycle G0/G1 increased(P=0.0011),and the S-phase decreased(P=0.0006)Cell cycle arrest was in the G0/G1 phase.The scratch closure of the ACV treatment group was obvious,and the closure rate was significantly reduced(P<0.05).As the ACV concentration increased,the inhibitory effect was more significant.Conclusion:Acyclovir inhibits the proliferation of HTFs and promotes apoptosis by blocking the cell cycle of HTFs;it is also involved in inhibiting the migration of HTFs cells.
