Proliferative vitreoretinopathy(PVR),ocular injury,postoperative complications,detachment of retina and so on ,they often concurred with ophthalmic proliferation,caused ophthalmic fibroplasia and formed fibrous membrane which acutely damage vision. Operation can't prevent but can stimulate cells of eyes to migrate and proliferate, caused ophthalmic fibroplasias. So it's very important significance to find new medicines for preventing and curring ophthalmic proliferation.Scar formation after operation of ophthalmic proliferation cause enormous suffering for patients. Follicular scar formation after glaucoma filtering operation is the main cause mostly affect the success rate. In the postoperative healing process, because of tissue wound, inflammatory reaction by operation and release of multifold cytokine, fibroblasts activate, migrate, proliferate and synthesize and secrete multifold extracellular matrix, and cause fibers proliferate in excess, and follicle cicatrizate at last. At this time, the filtering operation is defeated. Recently medication resisting metabolize such as mitomycin, fluorouracil has used to rise operation success rate, but it may cause severe complication, such as shallow anterior chamber, cataract, choroidal detachment and so on. So its application in clinic is confined.Correct appreciation and application of medication resisting cicatrisation, searching for absorbing completely, effective, low-toxicity new medicine insuppression of cicatrisation after glaucoma filtering operation are the important turning point of rising operation success rate.Quercetin (QU,3,4,5' ,3' ,4' - pentahydroxyflavone) and its derivant is the broadest distributed flavonoid in vegetable kingdom, has many kinds of biological activity and very high medicinal value, also is the most main flavonoid in the human diet. there are almost 100 kinds of Chinese traditional medicine which include quercetin in vegetable kingdom, it is extensively exisiting in many kinds of plants' leaf? the flower?the fruit, and the content of quercetin and its derivant in onions?apples and tea is respectively 28~49 mg? 2.1~7.2 mg? 1.7~2.5 mg per 100 grams. In recent years the physiological property of quercetin aroused the domestic and foreign scholars' widespread interest. They did many reseach work on its biological activity, discovered that quercetin has many pharmacologic action, such as expanding coronary artery, reducing vasopermeability, anti-virus, antiinflammation, anticancer, resisting hematoblast conglomeration, anti-anaphylaxis, relieving pain, antioxidation, scavenging free radical and so on. The best noticeable characteristic is resisting cell proliferation activity. It not only can resist lung?liver?kidney?heart and the eye fibrosis, but also inhibit the skin keloid formation. Quercetin as a natural flavonoid, not obviously adverse effect, widely resource, is a quite ideal new medicine of preventing scar formation in the glaucoma filtration surgery.Investigation of the effect of quercetin on the proliferation of human Tenon′s capsule fibroblasts is benefit for providing reliable theory evidence for quercetin as a new medicine of preventing scar formation in the glaucoma filtration surgery. Cultured fibroblasts were exposed to different concentrations of QU solution (20?40?60?80?100μmol/L). The proliferation of human Tenon′s capsule fibroblasts was detected by CCK-8 agent. We observed cells in invert microscope. Result indicates there is significantly difference between experimental group and control group in absorbance (20μmol/LgroupP<0.05, 40~100μmol/LgroupP<0.01). Quercetin can prohibit the growth of fibroblast of human Tenon′s capsule in vitro and show a dose-time dependent tendency. When the concentration of quercetin is 100μmol/L, the proliferation of human Tenon′s capsule fibroblasts is completely inhibited. The normal fibroblast of human Tenon′s capsule are attached cell, intensive fusiform, its cell body is slender and lucency under invert microscope. Growth of human Tenon′s capsule fibroblasts is good under microscope after Giemsa's staining method. The cell density is intensive and caryotin is uniformity. After cells were treated by 50μmol/L QU for 48 hours, we observed that cells were shrink, karyopycnosis, caryotin concentrated and aggregated to nuclear membrane edge. After cells were treated by 100μmol/L QU for 48 hours, the proliferation of human Tenon′s capsule fibroblasts wasn′t seen. We can observe nuclear concentrated, apoptotic body increased, partial rupture of membrane. Thisexperiment proved that quercetin can prohibit the growth of fibroblast of human Tenon′s capsule in vitro and show a dose-time dependent tendency. When the concentration of quercetin is 100μmol/L, the proliferation of human Tenon′s capsule fibroblasts is completely inhibited.Quercetin as a natural, extensive pharmacologic action flavonoid, is not only effective?low-toxicity, but also widely resource .It is a quite ideal new medicine of preventing scar formation in ophthalmic proliferation and the glaucoma filtration surgery. Therefore, the further study on pharmacologic action mechanism and the transformation of QU into clinical medication are extremely necessary and extensively prospect.
