ObjectiveTo investigate the relationship between expression of PITX3 and Parkinson's disease.We tested the expression of PITX3 in Parkinson's disease patients and rat models of Parkinson's disease to explore its effect on the pathogenesis of Parkinson's disease and PITX3 in different Hoehn-Yahr grade of Parkinson's disease patients further to determine the relationship between PITX3 and grade of Parkinson's disease.MethodTo examine this question,we use enzyme linked immunosorbent assay(ELISA)analysis detected the expression levels of PITX3 in each experimental rats.The experimental rats are divided into five groups: normal rats,3d PD group,7d PD group,14 d PD group and 28 d PD group.The PD rats model were induced by 6-OHDA injections.We used ELISA analysis detected the expression levels of PITX3 in 30 Parkinson's disease patients and in 30 healthy persons.According to gender,age,Hoehn-Yahr classification,course of disease,UPDRS?,we divided PD patients into different groups to compare the different level of PITX3.Statistical analysis:All statistical analysis was performed with Graphpad Prism and SPSS 21.0.T-test was used to explore differences between groups.Significance was declared when P<0.05.ResultsAnimal experiment:The ELISA tests demonstrated that the expression of PITX3 in PD group were reduced than in the normal group.Compared with the normal rats group the ELISA tests shows the levels of PITX3 declined in PD groups.As time goes on,the level of PITX3 decreased significantly.The results were statistically significant.(P < 0.05).Clinical trial:The ELISA tests demonstrated the expression levels of PITX3 was significantly lower than the control group(P<0.05).There was no significant difference in the level of PITX3 in different gender and age(P>0.05).The content of PITX3 in H-Y ?II group is higher than that in H-Y>II and there is a significant difference(P<0.05),with statistical significance.The PD group was divided into three groups based on the duration of the disease,the expression level of PITX3 in the duration of<5years group was significantly lower than the duration of 5-10 years group and ?10 years group(P < 0.05).However there is no significant difference between 5-10 years group and ?10years group(P>0.05).There was no significant correlation between PITX3 level and UPDRS-III score(P>0.05).Conclusion1.Parkinson's disease is related to the decrease of PITX3 in peripheral blood.PITX3 may play a role in the pathogenesis and progression of PD patients.2.The level of PITX3 in peripheral blood cell can be used as a biomarker for the diagnosis of PD.3.The expression level of PITX3 in peripheral blood in patients in the medium and late stage was significantly lower than that in early stage.4.The changes of level of PITX3 in the peripheral blood of patients with PD disease may show the changes of central dopaminergic neuron.
