Objective:The effects of platelet-rich concentrates on the biological characteristics of rabbit condylar chondrocytes in inflammatory microenvironment were studied by constructing an in vitro inflammatory model of condylar chondro-cytes.Methods:Rabbit primary condylar chondrocytes were cultured in vitro by enzyme pre-digestion tissue block adherence method.The third-generation rabbit condylar chondrocytes were stimulated with 10 ng/ml IL-1? for 24h,then cultured in DMEM containing 10%platelet-rich concentrate(PRP and I-PRF)for 24h.The effect of platelet-rich concentrate on the proliferation of rabbit condylar chondrocytes in inflammatory microenvironment was detected by CCK-8.The effect of platelet-rich concentrate on the biological characteristics of rabbit condylar chondrocytes in inflammatory microenvironment was detected by RT-PCR..Results:The results of CCK-8 showed that:(1)PRP and I-PRF significantly promoted the proliferation of rabbit condylar chondrocytes(P<0.05),but no statistical difference was found between PRP and I-PRF(P>0.05).(2)PRP and I-PRF significantly promoted the proliferation of rabbit condylar chondrocytes in the inflammatory microenvironment(P<0.05),but no statistical difference was found between PRP and I-PRF(P>0.05).The results of RT-PCR showed that:(1)PRP and I-PRF significantly inhibited the expression of IL-1? and MMP-13 in rabbit condylar chondrocytes(P<0.05),while the expressions of Aggrecan,Co12al and SOX9 were significantly increased(P<0.05),but no statistical difference was found between PRP and I-PRF(P>0.05).(2)PRP and I-PRF significantly inhibited the expression of IL-1?and MMP-13 in rabbit condylar chondrocytes under inflammatory microenvironment.(P<0.05),while the expression of Aggrecan,Col2al and SOX9 was significantly increased(P<0.05),but no statistical difference was found between PRP and I-PRF(P>0.05).Conclusion:(1)In vitro inflammatory model of condylar chondrocytes can be successfully established by using IL-1? to culture condylar chondrocytes;(2)PRP and I-PRF can promote the proliferation of rabbit condylar chondrocytes;(3)PRP and I-PRF inhibited the destruction of rabbit condylar chondrocytes in the inflammatory microenvironment.(4)There was no significant difference between PRP and I-PRF in promoting the proliferation of chondrocytes or inhibiting the inflammatory microenvironment. |
