| Objectives:This paper is to study the changes of inflammatory factors in the meibomian gland before and after eyelid surgery and the morphological structure changes of meibomian gland.Therefore,the effect of eyelid surgery on structure and function of meibomian gland can be explored.Methods:There were 22 adult clean New Zealand white rabbits with a weight of about 2-2.5kg,both male and female.The eyelid tissue of 15 mm long and 1 mm wide was removed along the rabbit’s upper eyelid margin.Their tears were extracted separately before surgery,3 days after surgery,1 week after surgery,2 weeks after surgery and 4 weeks after surgery for ELISA detection.The contents of EL-6,IL-8 and TNF-α were observed at different stages before and after surgery.And meibomian gland biopsy was performed before surgery,1 week after surgery and 4 weeks after surgery to observe the morphological changes and inflammation of the meibomian gland tissue.Results:(1)The results of ELISA showed that the levels of IL-6,IL-8 and TNF-a increased significantly at 3 days after surgery compared with those before surgery,and this difference has statistical significance(P<0.05).The levels of IL-6,IL-8 and TNF-α gradually decreased at 1 week,2 weeks and 4 weeks after surgery compared with those at 3 days after surgery,but the levels of inflammatory factors at 2 weeks after surgery were still higher than those before surgery,and the levels of IL-6 and IL-8 at 4 weeks after surgery were close to those before surgery,whereas the level of TNF-α was still higher than that before surgery.(2)Histopathological examination results:one week after surgery,the glands of the meibomian gland were evenly distributed,the acinar tissue shrank,the number of acinar cells was reduced,the morphology of some acinar cells was blurry.,the basic cells were unevenly distributed,the glandular duct cells were reduced,the glandular duct cavity was atrophied and inflammatory cell infiltration was seen around the meibomian gland.At 4 weeks after surgery,the distribution of glands in meibomian gland was not uniform,the size of acinar tissue was different and the arrangement was irregular as well as not all distributed along the ducts.Besides,the hyperplasia of acinar cells was obvious and some of them had blurry morphology.Meibomian gland ducts were unevenly distributed and some of them obviously expanded and narrowed.And a large number of new blood vessels could be seen around these ducts.Conclusions:(1)Inflammatory factors of meibomian gland have increased significantly after eyelid surgery.Particularly,TNF-alpha is the most important inflammatory factor that affects the function of meibomian gland among the three inflammatory factors.Therefore,eyelid surgery has induced the occurrence of MGD.(2)Morphology and structure of eyelids have changed significantly after surgery. |
