Objective:In recent years,amiodarone,the most commonly used drug with better effect,is widely used in clinical practice.Amiodarone is a kind of III antiarrhythmic drug.It is a multichannel blocker.It can block the Na channel and slow down the slow and slow K current?Iks,Ikr?at the same time.The tremor was treated effectively.As a pure Chinese medicine preparation,the main composition of Wenxin Granule is composed of 5 flavors of traditional Chinese medicine,Dangshen,Huangqi,Sanqi,amber and sweet pine.It has the effect of inspiring heart,supplementing blood and so on,making the dispelling of evil spirits,the spirit of an ideal and the smooth gas and blood,is a new generation of Chinese medicine which treat Arrhythmia,and has the effect of inhibiting many kinds of ion channels.This study will be based on the research status of atrial fibrillation,design and use the biological system and the ultrahigh phase liquid chromatography and mass spectrometry combined with the metabolic group technology platform,aiming at the treatment of atrial fibrillation rats with amiodarone combined with amiodarone combined with amiodarone to treat atrial fibrillation rats.To find out the differential characteristic metabolites and metabolic pathways of these two different treatments for the treatment of atrial fibrillation,and to analyze and verify whether the effect of amiodarone in combination with amiodarone is better than amiodarone alone,and a useful attempt is made to treat atrial fibrillation in these studies.The treatment provides new perspectives and ideas for further research on atrial fibrillation,and facilitates subsequent follow-up studies.Methods:24 Wistar rats were randomly divided into 4 groups,namely,atrial fibrillation group?AF Group?,single amiodarone group?Amio Group?,amiodarone group?Amio+WX Group?and blank control group?Blank Group?,with 6 rats in each group.Atrial fibrillation group,single amiodarone group and Wenxin Granule Combined with amiodarone group were given a certain proportion of Ach-Cacl2.A rat model of atrial fibrillation has been constructed by continuous injection of caudal vein for 7 days,once a day.After successful establishment of the model,amiodarone was given to rats in amiodarone group.Gavage 28 days,once a day,Wenxin Granule and amiodarone were given to rats in Wenxin Granule Combined with amiodarone group.Gavage for 28 days,once a day.After the end of the drug treatment,the atrial fibrillation group,the amiodarone group and Wenxin Granule combined with amiodarone group were used to record the changes of electrocardiogram?ECG?in the amiodarone group.After the intervention,1ml was taken from the inferior vena cava at the 10 day,20 days and 30 days after the drug intervention.The specimens were treated strictly according to the specimen of metabonomics,the model of rat atrial fibrillation metabolic profile was constructed,and the basic experimental data collected by the metabonomics platform were used to analyze the basic analysis using the Mzmine2.0 software.The principal component analysis?PCA?of SIMCA-P+12.0.1.0 software is used to obtain the principal component analysis model?PCA model?and the orthogonal minimum bias two multiplying model?OPLS-DA?to further screen the characteristic metabolites.The metabolites were introduced into the Metaboanalyst 3 database to analyze metabolic pathways,and further explore metabolic pathways that are highly correlated with atrial fibrillation therapy.Results:In the results of this study,the duration of atrial fibrillation in the amiodarone group and the amiodarone group was significantly lower than that of the atrial fibrillation group after the drug treatment.The difference was statistically significant in comparison with the atrial fibrillation group.This study successfully constructed the model of atrial fibrillation rat.After the prognosis of drug drying,the principal component analysis model?PCA model?was successfully constructed.The PCA model was a model of 8 principal components?R2X=53%,Q2=29%?,and the correction model was successfully constructed,that is,the orthogonal partial least squares model?OPLS-DA model?,and the OPLS-DA model with 7.For predicting principal components,7 orthogonal principal components have good goodness of fit?R2X=41.2%,R2Y=25%,Q2=6.14%?.There were 9 kinds of Characteristic Metabolites obtained by p<0.01,4-Hydroxy-L-proline,Stearoylcarnitine,Polyoxyethylene?600?monoricinoleate,L-2-Amino-4-methylenepentanedioic acid,LysoPC?O-18:0?,Nonadecanoic acid,LPA?P-16:0e/0:0?,Glycocholic acid,Glycerophosphocholine.The metabolic pathway obtained by Metaboanalyst software are Ether lipid metabolism?Glycerophospholipid metabolism and Primary blie acid metabolism.Conclusion:This experiment successfully constructed a rat model of atrial fibrillation.From the results of electrocardiogram statistics and metabonomics,it was proved that the effect of amiodarone on the effect of amiodarone was more effective than amiodarone alone.Lu,from the point of metabonomics,describes the mechanism of the treatment of atrial fibrillation with the combination of Wenxin Granule and amiodarone,and provides a new reference and idea for further experiments to reveal the pathogenesis of atrial fibrillation and the mechanism of drug therapy for the treatment of atrial fibrillation.
