Metabonomics, including discovery metabonomics based on non-discrimination analysis and target metabonomics based on targeted quantitative compounds, was widely applied in clinical diagnosis, toxicology and other fields. This study aimed to develop a metabonomic method based on liquid chromatography-mass spectrometry and to explore its application in complex system such as traditional Chinese medicine and complex diseases.Based on the technique of UPLC/TOF-MS, the efficacy and mechanism of the clinical effective Shuanglong Formula and NSLF6 with comparatively clear phytochemical composition, comparatively clear mechanism and controllable quality were studied with the guidance of Chemomics by the isoproterenol-induced and coronary artery ligation induced myocardial infarction in rats. Metabonomic results combing with physiological indicators suggested that NSLF6 could treat MI via several mechanisms such as speeding up the rehabilitation and reconstruction of the heart by promoting cardiomyocytes renewal and promoting angiogenesis, improving the energy metabolism by adjusting the citric acid pathway, and promoting anti-myocardial ischemia by decreasing the myocardial injuries from the glycolysis products.The acute toxicity of toad venom on rat heart was investigated with the guidance of toxicology and metabonomics. The results showed that the disturbed metabolisms by toad venom toxicity were concentrated on lipids related pathways, reflecting the heart damage caused by toad venom from the metabolic level. We could find that the Chinese medicine Shexiangbaoxin tablet could reduce or eliminate the toxicity of toad venom from the angle of compatibility of Chinese medicine. Metabonomics analysis and the ECG results had a good correlation in this study. This study can provide a reference for the study of mechanism of toad venom toxicity and clinical use of drugs.A method for the simultaneous determination of 16 metabolites in folate, homocysteine and glutathione metabolism was established, and applied to the investigation of neural tube defects (NTDs). Based on the above analysis, the metabolic findings combining with statistical analysis showed that 7 potential biomarkers associated with embryonic development were found out. The NTDs pathogenesis was discussed. At the same time, the ROC curve was used to evaluate the accuracy of clinical diagnosis of the 7 potential biaomarkers and found that 5-methyltetrahydrofolate may be an important indicator in the cilinical diagnosis. |
