Cancer is a serious threat to human health,one in every three diagnosed cancer cases is skin cancer.There are more than 2 million cases of skin cancer in the world every year,the death rate of malignant melanoma is very high.Ultraviolet radiation is an important factor in the development of skin cancer.With the destruction of the ozone layer,more and more ultraviolet rays reach the ground.Humans are exposed to ultraviolet light for a long time,especially UVB,causesing skin aging,DNA damage,photoaging,and even cancer.At present,the study of the anti-tumor and anti-UV effects of ginseng active ingredients have become a hot spot.But the heavy metals,pesticides and solvent residues in ginseng and its extracts pose safety risks to human.Ginseng grows slowly and occupies a large area of forest land.In order to solve the problem of cut trees to plant ginseng,the researchers used ginseng stems and leaves as explants to induce ginseng hairy roots to the large-scale production of ginsenoside in the factory.The studies on the ginseng hairy roots were mainly focused on the induction of ginseng hairy roots,the optimization of culture conditions and the production of secondary metabolites.However,the study of effects of ginseng hairy roots on anti-melanoma and anti-UV radiation has not been found.So we chose ginseng hairy roots and its culture medium which are rich in ginsenosides,polysaccharides,amino acids,without pesticides,heavy metals and stable quality as raw materials.Firstly,the effective components were determined by HPLC and phenol-sulfuric acid method.Secondly,three methods of Cu2+chelating ability,PPO and FRAP were used to evaluate the antioxidant effect of ginseng hairy roots.Thirdly,the anti-melanoma activity of ginseng hairy roots in vitro was studied.Finally,according to the results of efficacy evaluation,we developed a skin care lotion of ginseng hairy roots to study its anti-ultraviolet effect and mechanism and provide theoretical basis to reference for the further study and application of ginseng hairy roots pharmacology.The experimental results are as follows:1.The antioxidant capacity of ginseng hariy roots is remarkable.With the increase of concentration,the Cu2+chelating rate of ginseng hariy roots is approaching VC.At the concentration of 100 mg/mL,there was no significant difference between the Cu2+chelating rate and VC in the ginseng hariy roots?P>0.05?;ginseng hariy roots and VC have the same scavenging trend of enzyme activity;at the concentration of 40-100mg/mL,the difference was not significant?P>0.05?.The total antioxidant capacity of the ginseng hariy roots reached0.8774 mmol FeSO4/g at 100 mg/mL.2.Inhibition of proliferation of melanoma A375 cells by ginseng hairy roots?P<0.05?;inhibition of tyrosinase activity in A375 cells?P<0.05?;reduce the production of melanin in cells;reduce cell migration and invasive ability.Compared with the normal control group,there were obvious apoptotic cells in the ginseng hairy roots group,ginseng group and dacarbazine group.Compared with the ginseng group and dacarbazine group,the ginseng haiy roots group has higher apoptosis rate in the late stage?P<0.05?.This effect achieved by blocking the cell cycle of skin cancer in S and G2/M phases.Ginseng hairy roots promotes the proliferation of human keratinocyte HaCaT and cell scratch healing.The protective effect of ginseng hairy roots to HaCaT cells is stronger than ginseng?P<0.05?.However,dacarbazine produces generate cytotoxicity to HaCaT cells and inhibited cell proliferation.3.Preparation of ginseng hair root lotion:Mixing 90 mL deionized water with 5 mL glycerol and 5 mL propylene glycol,then 0.1 g sodium hyaluronate was added,and 0.1 g carbomer was added slowly under high speed.10%sodium hydroxide was used to regulate the pH.The formula ginseng hairy roots lotion is stable.4.In the ginseng hairy roots groups,the Hyp content and GSH-Px activity in the skin were increased,MDA content was decreased,CAT,SOD vitality were increased?P<0.05?.The IL-6and TNF-?content in the ginseng hairy roots groups were decresed?P<0.05?.The ginseng group,middle dose group of hairy roots and high dose group of hairy roots effectively improved skin injury and desquamation of UVB-damaged mice skin,reducing the thickness of mice epidermis,reducing inflammatory infiltration and hyperkeratosis of the stratum corneum,protecting collagen fibers.The expression of MMP-1 and MMP-3 protein in the skin of model group was significantly higher than that in normal control group?P<0.05?.The expression level of MMP-1,MMP-3 protein were decreased significantly in the ginseng group,low,medium and high dose group of ginseng hariy roots?P<0.05?.
