Objective: To investigate the effect of platelet-rich fibrin on the expression of matrix metalloproteinase-9(MMP-9)in wounds of diabetic rats and the mechanism of promoting wound healing.Methods: Total of 40 normal male rats were selected.The rats were fed with high-fat and high-sugar diet and injected with streptozotocin to prepare diabetic rats.The rats with successful modeling were screened.After anesthesia,a square with a length of 22 mm was cut in the back symmetrical part.The left side wounds were disposed with platelet-rich fibrin(PRF)(PRF group),and the right wounds were disposed with Vaseline gauze(control group).The wound healing was observed on both sides of the rats.Masson staining were performed to observe the change of collagen.SP immunohistochemistry was used to detect the expression of MMP-9 during wound healing.Results: The wound healing in the PRF group was significantly better than that in the control group on the 7th and 14 th after surgery(68.7%±10.6% vs 46.6%±9.2%,83.7%±2.2% vs 72.1%±3.4%,both P<0.05).Masson staining showed that the collagen in the PRF group increased significantly compared with the control group on the 7th and 14 th day after surgery(47.6%±6.3% vs 29.4%±6.5%,77.1%±5.7% vs 44.2%±7.8%,both P<0.05).SP immunohistochemistry showed that the expression level of MMP-9 in PRF group was significantly lower than that in the control group on the 3rd,7th and 14 th day after surgery(37.52±2.03 vs 39.83±1.56,22.59±1.53 vs 29.28±2.15,10.28±2.44 vs 17.39±2.08,both P<0.05).Conclusion: PRF can accelerate the healing of wounds.The mechanism may be related to inhibiting the expression of MMP-9,thereby reducing collagen degradation in the wound.inhibit the expression of MMP-9 in wounds of diabetic rats and...
