Purpose: To explore the expression profile variation of circular RNA?circ RNA?in peripheral blood of subjects with non-arteritic anterior ischemic optic neuropathy?NAION?and without NAION,and to analyze the differential expression results,and predict the role of circ RNAs in disease development,providing new ideas and new methods for treatment and diagnosis.Method: Applied High-throughput sequencing to explore the expression profiles of RNAs in peripheral bloods of 6 NAION patients and 5 healthy controls.quality control Obtained the advanced data the original data by through ticking out the unqualified data.Then performed cluster analysis,volcano blot,co-expression network,Gene ontology?GO?analysis and Kyoto Encyclopedia of Genes and Genomes?KEGG?pathway to analyse the whole expressed genes.Lastly,using the quantitative real-time Polymerase Chain Reaction?q RT-PCR?to verify those significantly differentially expressed circ RNAs and do some bioinformatics analysis and prediction in 12 NAION patients and 12 controls.Results: High-throughput sequencing showed that there were significant differences in the expression of 49 circ RNAs and 1161 m RNAs in peripheral blood of NAION patients.in which there were 24 circ RNAs upregulation and 25 circ RNAs downregulation?variation folds > 2 and P < 0.05?.Real-time quantitative PCR confirmed that hsacirc0005583,hsacirc0003922,hsacirc0002021 and hsacirc0000462 were significantly downregulated in peripheral blood of NAION patients?variation folds> 2 and P < 0.05?,especially hsacirc0005583 was the most significantly downregulated circ RNA?P < 0.001?.These differentially expressed circ RNAs were predicted findings may be related to processes such as neurodegeneration,oxidative stress,immunity,and metabolism.Conclusion: The expression profile of circ RNAs in peripheral blood of NAION patients is significantly changed.Differentially expressed circ RNAs may provide a new idea for the cause of the disease.
