Objective: This research aims to screen differentially expressed circular RNAs in human osteosarcoma tissues using high-throughput sequencing technology and bioinformatics methods.Methods: Five osteosarcoma tissues?experimental group?in the center of the disease detection tissue before neoadjuvant chemotherapy were selected from 5 patients with osteosarcoma,and 5 normal cartilage tissues?control group?were selected to extract total RNA.Then,through high-throughput sequencing technology and bioinformatics methods,screen the differentially expressed circular RNA,predict microRNA,and then classify and enrich the KEGG biological pathway for genes derived from the differential circular RNA.Results:1.A comparative study of the experimental group and the control group found that among the differentially expressed circular RNAs screened,61,337 circular RNAs were up-regulated and 7,132 were down-regulated,and 59,655 circular RNAs were only expressed in human osteosarcoma tissue in the experimental group There were 3,316 circular RNAs expressed only in the normal cartilage tissue of the control group,and 5498 circular RNAs co-expressed in the two groups.2.The study found that hsacirc0001445,hsacirc0006988,and hsacirc0000246 were significantly up-regulated in human osteosarcoma tissue,while hsacirc0009043,hsacirc0072309,and hsacirc0001185were significantly down-regulated.Enrichment analysis was performed on the source genes of circular RNA,and 49 signaling pathways were screened withsignificant differences.Conclusion:1.In the 5 cases of human osteosarcoma tissue in this study,a total of 6significantly differentially expressed circular RNAs were screened,namely hsacirc0001445,hsacirc0006988,hsacirc0000246 were significantly up-regulated,hsacirc0009043,hsacirc0072309,hsacirc0001185 were significantly down-regulated.2.This study found that ubiquitin-mediated proteolytic signaling pathway,AMPK signaling pathway,sphingolipid signaling pathway and insulin signaling pathway are closely related to the pathogenesis of osteosarcoma,and the phosphatidylinositol signaling pathway may be related to the metastasis of human osteosarcoma.
