Purpose: In this study,non-arteritic anterior ischemic optic neuropathy patients and healthy normal human blood samples were subjected to high-throughput sequencing and their transcriptome data were analyzed.Differentially expressed LncRNAs were screened and their expression profiles were analyzed,the relationship between the development of Lnc RNA and NAION is studied in order to find LncRNA that can be used as a diagnostic and therapeutic target for NAION.Method: RNA was extracted from peripheral blood cells of 6 NAION patients and 5healthy control groups,and transcriptome sequencing was performed using high-throughput sequencing to screen out differentially expressed Lnc RNAs.The resulting LncRNAs were analyzed for transcriptome expression profiling and bioinformatics Methods Functional analysis,cluster analysis and biological prediction of differentially expressed LncRNA were performed.According to the bioinformatics results,the expression of Lnc RNA screened was further verified by qRT-PCR technology.Results:(1)Through high-throughput sequencing,996 Lnc RNAs with significant differences in peripheral blood were found in NAION patients.317 LncRNAs were up-regulated and 679 LncRNAs were down-regulated(variation folds > 2 and P <0.05).(2)Through GO database analysis,differentially expressed LncRNAs are involved in various biological processes related to NAION,such as apoptosis,lipid metabolism,MAPK signaling pathway,inflammatory response,and oxidative stress.(3)The qRT-PCR method was used to verify the expression of the six LncRNAs at the cellular level,and it was found to be consistent with the results of gene chip detection.The results show that LncRNA can be used as a biomarker and therapeutic target in the clinical diagnosis and treatment of NAION and drug development.Conclusion:(1)Differentially expressed LncRNAs in peripheral venous blood of NAION patients were screened to establish NAION-related Lnc RNA differential expression profiles.(2)It predicts that differentially expressed LncRNAs are involved inthe regulation of multiple signaling pathways in the occurrence and development of NAION,and provide new clues for the research of NAION diagnosis and treatment.
