Background: To explore the therapeutic effect of traditional Chinese medicine Tongbi prescription on phlegm-dampness CIA mice,and to clarify the effect of expectorant dehumidification method on serum leptin and IL-17 in phlegm-dampness type CIA mice.On the basis of the phlegm-dampening CIA mice,the Tongbi prescription was administered to the stomach.By interpreting the severity of the joints of the mice and the detection results of specific indexes in the serum,the Tongbi prescription was explored in the phlegm-dampening CIA.It provides an objective basis for further study of the pathogenesis and clinical treatment of rheumatoid arthritis.Materials and Methods: A total of 34 SPF 4-week-old DBA/1 male mice were randomly divided into two groups.one group was blank group,6 mice were fed with normal diet,and the remaining 28 mice were fed with high-fat diet.After 7 weeks of continuous feeding,6 of 28 mice were randomly selected as phlegm-dampness group,and the remaining 22 mice were modeled by CIA.After the completion of the model,22 mice were randomly divided into phlegm-dampness CIA group(n = 6),traditional Chinese medicine group(n = 8)and western medicine group(n = 8).The traditional Chinese medicine group and the western medicine group were given Tongbi prescription and western medicine Rifampicin Tablets.Finally,they were divided into blank group(n = 6),phlegm-dampness group(n = 6),phlegm-dampness CIA group(n = 6),traditional Chinese medicine group(n = 8)and western medicine group(n = 8).The mice were killed 6 weeks after CIA modeling.During the experiment,the general situation of mice in each group was observed,and the incidence of arthritis in mice was scored.After extraction of materials from experimental mice,the pathological changes of knee joint tissue were observed and evaluated by HE staining,and the concentrations of blood lipid,IL-17 and leptin in serum were detected by enzyme-linked immunosorbent assay(Elisa).Results: 1.At the 1st-7th week,the mice in the blank group were in good condition and had no symptoms of phlegm-dampness;the mice in the phlegm-dampness group were slow to react and squinted;tired,sleepiness and pile-up;obese in shape,dim in hair,yellow and lustrous in color;and the feces were not shaped and light in color.At the 8th to 11 th week,after the establishment of CIA model,the mice in phlegm-dampness CIA group showed the aggravation of phlegm-dampness symptoms after the establishment of the model.At the 12th-14 th week,after administration of traditional Chinese medicine group and western medicine group respectively,the symptoms of mice in the two groups were relieved,especially in the traditional Chinese medicine group.2.From week 1 to week 7,the body weight of mice in phlegm-dampness group and phlegm-dampness CIA group was higher than that in blank group.At the 8th to 11 th week,after CIA modeling,the body weight of mice in phlegm-dampness group and phlegm-dampness CIA group showed a continuous upward trend.From the 12 th to 14 th week,the body weight of mice in the traditional Chinese medicine group and the western medicine group showed a downward trend.3.At the 8th to 14 th week,the AI score of mice in blank group and phlegm-dampness group was 0.From the 11 th to 13 th week,the joint AI score of mice in phlegm-dampness CIA group,traditional Chinese medicine group and western medicine group continued to increase.From the 13 th to 14 th week,the joint AI score of mice in phlegm-dampness CIA group continued to increase,and the joint AI score of mice in traditional Chinese medicine group and western medicine group decreased,especially in traditional Chinese medicine group,and the difference was statistically significant compared with western medicine group(P < 0.05).4.The pathological sections of the joint of the mice in the blank group showed that the structure of the knee joint was normal,there was no synovial tissue proliferation,no inflammatory cell infiltration,no pannus hyperplasia,no articular cartilage erosion,and the articular surface was smooth.Compared with the blank group,there were a small number of inflammatory cell infiltration,no synovial tissue proliferation,no pannus hyperplasia and articular cartilage erosion and destruction in the phlegm-dampness group.Compared with the blank group,the articular surface of mice in phlegm-dampness CIA group was rough,synovial tissue proliferated,a large number of inflammatory cells infiltrated,and the joint space was covered with pannus.Compared with phlegm-dampness CIA group,synovial tissue proliferation,inflammatory cell infiltration,pannus and bone destruction were decreased in traditional Chinese medicine group and western medicine group,especially in traditional Chinese medicine group.5.The levels of serum TC,TG and LDL in the phlegm-dampness CIA group and the phlegm-dampness group were significantly higher than those in the blank group(P<0.05),and the HDL level was significantly lower than that in the blank group(P<0.05).The levels of TC,TG and LDL in the Chinese medicine group were significantly lower than those in the phlegm-dampness CIA group(P<0.05),and the HDL level was significantly higher than that in the phlegm-dampness CIA group(P<0.05).6.The levels of IL-17 and leptin in phlegm-dampness CIA group were significantly higher than those in blank group,phlegm-dampness group,traditional Chinese medicine group and western medicine group(P< 0.05).The level of traditional Chinese medicine group was significantly lower than that of western medicine group,the difference was statistically significant(P < 0.05).7.There was a positive correlation between serum TC level and leptin,the correlation coefficient was r=0.599,P<0.001.There was a positive correlation between leptin and IL-17,the correlation coefficient was r=0.454,P=0.008.Conclusions: 1.Tongbi prescription can alleviate the symptoms of phlegm-dampness in CIA mice with phlegm-dampness type.2.Tongbi prescription can improve the arthritis index and the degree of joint lesion in phlegm-dampness CIA mice.3.Tongbi prescription may inhibit the secretion of IL-17 by inhibiting leptin,so as to realize the therapeutic effect on phlegm-dampness CIA mice.
