Correlation Between PNPLA3 Rs738409 SNP And HCC In Han Population Of Northeast China

Objective: There have been a large number of studies on the relationship between PNPLA3 rs738409 SNP and HCC in foreign countries,but there is still a large sample case-control study on the correlation between PNPLA3 rs738409 SNP and HCC in China.Therefore,we analyzed the PNPLA3 rs738409 SNP in HCC patients in Northeast China.To study the correlation between PNPLA3 rs738409 SNP and the incidence of HCC in Chinese Han population.And to explore whether PNPLA3 rs738409 SNP is a high risk factor for the onset of HCC.Methods: A case-control study was conducted to screen 217 HCC patients as a case group in the Han population of Northeast China.Among them,109 patients with hepatitis B after hepatitis B virus and 70 patients with liver cancer after hepatitis C virus,38 patients with liver cancer after alcoholic liver disease,164 normal people as a healthy control group.The case components were liver cancer group after hepatitis B virus,liver cancer group after hepatitis C virus and liver cancer group after alcoholic liver disease.The SNP of PNPLA3 rs738409 locus was detected by RT-PCR.Results:(1)Basic situation: There was no significant difference in gender and age distribution between healthy control group and case group(P = 0.895 and 0.344,respectively),which was comparable.The albumin of the case group was significantly lower than that of the healthy control group,and the liver enzymes,TBIL and AFP were significantly higher than the healthy control group.There were no significant differences in albumin,liver enzymes and TBIL between the groups except AFP(P>0.05).(2)Hardy–Weinberg genetic balance test: The allele frequency of rs738409 locus in healthy control group and case group accorded with Hardy-Weinberg equilibrium(healthy control group PNPLA3 rs738409 C>G:P=0.928,case group PNPLA3 rs738409 C>G: P = 0.088),in accordance with Hardy–Weinberg's law of genetic balance,the sample is representative of the population.(3)Gene distribution of PNPLA3 rs738409 between groups and groups: According to different fluorescent markers on the probe,PNPLA3 rs738409 gene was divided into three genotypes: GG,CG and CC,and CC,CG,in healthy control group.The frequencies of GG genotype and G allele were 0.482,0.427,0.091,and 0.305,respectively.Thefrequencies of CC,CG,GG genotypes and G alleles in the case group were 0.373,0.429,0.198,and 0.412,respectively.PNPLA3 The distribution of gene rs738409 genotype in the case group and healthy control group was statistically significant(P=0.004)and the frequency of PNPLA3 rs738409 G allele was significantly higher in the case group than in the healthy control group(OR =9.313,P=0.002).The frequencies of CC,CG,GG genotypes and G alleles in Hepatitis B cirrhosis group were 0.367,0.431,0.202,and 0.417,respectively.The genotype and allele distribution of healthy control group and hepatitis B cirrhosis group were statistically significant.(P = 0.007 and 0.009 respectively);the frequencies of CC,CG,GG genotypes and G alleles in hepatitis C cirrhosis group were 0.457,0.386,0.157,0.350,respectively,and the genotypes of healthy control group and hepatitis C cirrhosis group and There was no significant difference in allele distribution(P = 0.337 and 0.143,respectively).The frequency of CC,CG,GG genotype and G allele genotype in alcoholic cirrhosis group were 0.237,0.500,0.263,0.513,respectively.There were significant differences in genotype and allele distribution between the control group and the alcoholic cirrhosis group(P = 0.001 and 0.009,respectively).(4)Relationship between PNPLA3 rs738409 and biochemical indicators of each group: The case group was divided into CC/CG and GG groups according to whether or not the PNPLA3 rs738409 G allele was carried,and in the hepatitis B group,hepatitis C group and alcohol group,CC/CG and GG group.There were no significant differences in age,gender,liver enzymes,and TBIL differences.(5)Relationship between PNPLA3 rs738409 and cirrhosis grading: Patients with cirrhosis in the case group were divided into ?7 group and <7 group according to Child-Pugh score,Child-Pugh score ?7 group patient PNPLA3 rs738409 GG gene.The type ratio was significantly higher than the <7 group,and the difference was statistically significant(P=0.046).Conclusion: PNPLA3 rs738409 SNP is associated with the occurrence of liver cancer after liver cancer and alcoholic liver disease in Han population of Northeast China.
