| Objective:To examine the therapeutic efficacy of QDG against elevated blood pressure using the SHR model,as well as the mechanisms behind its anti-hypertensive activity and protection against cardiac inflammation.To further illuminate the underlying mechanisms of QDG in protecting against hypertension,provides a theoretical basis for the widespread use of QDG in the prevention and treatment of hypertension.Methods:1.Blood pressures of 1 8 male Spontaneously Hypertensive(SHR)rats and 6normotensive control Wistar Kyoto(WKY)rats were measured once per week,with the tail-cuff plethysmograph method using CODA? non-invasive blood pressure system.All rats were acclimatized for one week prior to experiments.After 8 weeks,the rat’s cardiac were eviscerated.2.Examined the histological changes in the cardiac of rats using Harris Hematoxylin and Eosin(H&E)staining.3.Real-time PCR detected the mRNA levels of MCP-1 and CCR2 in the cardiac.4.Immunohistochemical staining to analyze the expression of CD3,Mac-2,TNF-α,IL-6and IFN-γ,MCP-1,CCR2 in the cardiac.5.Elisa analysed the protein expression of TNF-α,IL-6 in the serum.Results:1.According to results of blood pressure,after treatment with QDG for three weeks has significantly attenuated the elevation of SBP,DBP,MAP in rats.2.H&E staining showed that there is a significant disorder in the arrangement of myocardial fibers,necrosis and Increased number of inflammatory cells in the myocardium compared to the normotensive WKY rats.However,the injures to the cardiac in SHRs was robustly mitigated by QDG treatment.3.Real-time PCR detected the mRNA levels of MCP-1 and CCR2 were all promoted inSHRs group,which was reversed following QDG treatment.4.Immunohistochemical staining suggested the high expressions of CD3,Mac-2,TNF-α,IL-6 and IFN-γ,MCP-1,CCR2 in SHRs,which was robustly attenuated following QDG treatment.5.ELISA showed that the protein expression of TNF-α and IL-6 were all increased in SHRs group,which was also attenuated following QDG treatment.Conclusion:1.QDG could significantly attenuate the elevation of blood pressure in an SHR model.2.QDG can improve the pathomorphology of SHR heart and alleviate the inflammatory reaction.3.QDG treatment also mitigated the cardiac inflammation via regulated the MCP-1/CCR2 signaling pathway. |
