Purpose:This experiment intends to use acupuncture at the "Zusanli" and "Sanyinjiao" points.Apply the therapeutic principles of invigorating the spleen and stomach,diuresis and lipid-lowering,taking the heart as the research object,the MCP-1/CCR2 signaling pathway is the entry point,observe the effects of acupuncture on the MCP-1/CCR2 signaling pathway and related inflammatory factors IL-6,TNF-αand cardiac inflammation.Materials and Methods:Select 10 6-week-old male Wistar-Kyoto rats(WKY rats)as the normal group,forty male6-week-old spontaneously hypertensive rats(SHR)were divided into model group,obesity model group,acupuncture group,and acupuncture control group according to the random number table method.One week later,the normal group and the model group were fed with ordinary feed.The obesity model group,acupuncture group,and acupuncture control group were fed with high-fat diet.Electroacupuncture was performed after successful model building.The rats in the normal group,model group and obesity model group are tied to a wire fence and fixed daily.No needling treatment.The acupuncture group uses the same fixation method as the first three groups,and acupuncture treatment.The acupuncture control group used the same fixation method as the other four groups.Choose a fixed acupuncture point at the upper part 1/3 of the rat’s tail,and the others are the same as the acupuncture group.After all treatments,serum and heart tissues of rats in each group were taken.Detect the content of IL-6 in rat serum by ELISA method;HE staining to observe the pathological changes of rat heart tissue;Detect the m RNA expression of CCR2,TNF-α,IL-6 by fluorescence quantitative Q-PCR method;Werstern blot method was used to detect the protein content of MCP-1 and CCR2.Result:1.The results of blood pressure monitoring show:Compared with the normal group,the blood pressure of rats in the model group,obesity model group,acupuncture group and acupuncture control group increased significantly(P<0.05);Compared with the obesity model group,the blood pressure of rats in the acupuncture group was significantly reduced(P<0.05).2.The weight monitoring results show:Compared with the normal group,the body weight of rats in the model group,obesity model group,acupuncture group and acupuncture control group increased significantly(P<0.05);Compared with the obesity model group,the body weight of rats in the acupuncture group was significantly reduced(P<0.05).3.The results of HE staining showed that the cardiac tissue structure of rats in the normal group was not abnormal;Rats in the model group and the obesity model group showed necrosis of myocardium and increased inflammatory cell infiltration;After treatment in the acupuncture group,the degree of myocardial necrosis was reduced,and the infiltration of inflammatory cells was significantly reduced.Compared with the acupuncture group,the rats in the acupuncture control group had obvious myocardial necrosis and increased inflammatory cell infiltration.4.Q-PCR results show:Compared with the normal group,the expression of CCR2 m RNA in the heart tissue of rats in the model group and the obesity model group increased significantly(P<0.05),the expression of TNF-αand IL-6 m RNA was significantly reduced(P<0.05);Compared with the model group and the obesity model group,the expression of CCR2 m RNA in the heart tissue of rats in the acupuncture group was significantly reduced(P<0.05),the expression of TNF-αand IL-6 m RNA increased significantly(P<0.05).5.Western blot results show:Compared with the normal group,the protein content of MCP-1 and CCR2 in the heart tissue of rats in the model group and obesity model group was significantly increased(P<0.05);Compared with the model group and the obesity model group,the protein content of MCP-1 and CCR2 in the heart tissue of rats in the acupuncture group was significantly reduced(P<0.05).6.ELISA results show:Compared with the normal group,the serum IL-6 content of rats in the model group and obesity model group was significantly increased(P<0.05);Compared with the model group and the obesity model group,the serum IL-6 content of the acupuncture group significantly decreased(P<0.05).In conclusion:1.Reduce the heart inflammation caused by hypertension and obesity,improve the heart disease of obese rats with hypertension,it is possible to treat the "Zusanli" and "Sanyinjiao" points with electroacupuncture,to achieve the purpose of inhibiting the MCP-1/CCR2 signaling pathway and the activities of related inflammatory factors IL-6 and TNF-α.
