Objective:To study on an animal model of sub-healthy nocturia of kidney-yang deficiency in accordance and provide theoretical basis for the intervention of traditional Chinese medicine on sub-healthy nocturia of kidney-yang deficiency.Methods:Thirty SD male rats were randomly divided into two groups:normal group and sub-healthy nocturia of kidney-yang deficiency group?model group?with 15 rats in each group after routine feeding at room temperature for 1 week.Normal group:Normal saline(20ml·kg-1·d-1,concentration 0.9%)was given intragastrically once a day for 10 days.Model group:Adenine(20ml·kg-1·d-1,concentration 1%)was given intragastrically once a day for 10 days.In the process of modeling,the changes of hair color,cold limbs,body type,mentality and feces were observed,the body weight was recorded every day,and the urine volume on the 8th,9th and 10th day was collected by metaboliccage.The concentrations of serum follicle stimulating hormone?follicle-stimulating hormone,FSH?and luteinizing hormone?luteinizing hormone,LH?in rats were measured by radioimmunoassay?radioimmunoassay,RIA?,and the pathological changes of kidney,testis and hypothalamus were observed.Results:1.General situation:Tthe rats in the model group appeared different degrees of mental malaise,physical emaciation,cold fear of cold limbs,like clustering,reduced activity,increased urine volume.The median?quartile spacing?of syndrome total score in normal group and model group was 32?32,32?and 29?26,32?,respectively,and the difference was statistically significant?P<0.05?.2.Indicators:There was a statistically significant difference in body weight between the normal group and the model group?246.85±19.48g VS 232.43±13.61g,P<0.05?.There were statistically significant differences in the quality of left kidney?1.25±0.12g VS 2.27±0.35g,P<0.05?andthe quality of right kidney?1.22±0.11g VS 2.36±0.39g,P<0.05?between the normal group and the model group.There was no significant difference in testicular mass?2.74±0.29g VS 2.77±0.21g,P>0.05?between the normal group and the model group.There were statistically significant differences in left kidney index?41.86±3.01mg/10g VS 91.71±16.49mg/10g,P<0.05?,right kidney index?40.73±2.60mg/10g VS 95.2±17.89mg/10g,P<0.05?and testicular index?91.41±5.28mg/10g VS108.79±9.29mg/10g,P<0.05?between the normal group and the model group.The median?interquartile spacing?of daytime urine volume?mL?,nighttime urine volume?mL?and all-day urine volume?mL?in the normal group were 3?1.5,4.2?,5?3,6.95?and 8.5?4.75,11.45?,respectively.The median?interquartile spacing?of daytime urine volume?mL?,nighttime urine volume?mL?and all-day urine volume?mL?in the model group were 13?11.75,14.5?,17?12.5,20.5?and 29.5?24,35?,respectively.There were statistically significant differences in urine volume in the normal group and the model group?P<0.05?.There were statistically significant differences in FSH concentration?4.20±1.31 mL VS 3.34±0.79 mL,P<0.05?and LH concentration?3.53±1.56 mL VS 2.44±1.30 mL,P<0.05?between the normal group and the model group.3.Pathological changes of kidney,testis and hypothalamus:naked eye observation:In normal group the appearance of kidney was bright red,the color of renal cortex and medulla was bright red when sectioned,and the structure was clear.In the model group,the volume of kidney was increased,leukoplakia was found on the renal cortex,and the color of renal cortex and renal medulla became lighter.There was no difference in testis and hypothalamus between the two groups.Microscopic observation:The structure of glomeruli and renal tubules was normal in the normal group.In the model group,the number of glomeruli decreased,the renal capsule cavity became larger,and the renal tubular cavity became larger.Testis:In normal group the morphology of testicular seminiferous tubuleswas regular,and the spermatogenic cells and sperm layers were arranged neatly.In the model group,the arrangement of spermatogenic cells and sperm levels was disordered and the levels were reduced.Hypothalamus:In the normal group,the neurons and glial cells were normal,the number of nerve cells in the model group was decreased,the nerve cellnucleis was large,the shape of some nerve cellnucleis was irregular,and the number of glial cells was increased.Conclusion:1.The sub-health model of kidney-yang deficiency was successfully established in the early stage.In this study,the nocturia model was established,and changes in renal tissue,hypothalamus tissue and testicular tissue were found.2.The animal model of TCM syndrome type of nocturia in this experiment is simple,which can create good conditions for the basic research of TCM intervention of nocturia.
