Systems Pharmacology Uncovers The Mechanism Of Tongqiao Biyan Formula For The Treatment Of Chronic Rhinosinusitis

Chronic rhinosinusitis(CRS)is a kind of chronic inflammation of nasal cavity and sinus mucosa with extremely high prevalence.Because of its synergistic effect and small side effects,traditional Chinese medicine(TCM)prescription has great advantages in the prevention and treatment of complex diseases.Tongqiao Biyan Formula(TBF),a traditional Chinese medicine prescription,can effectively relieve the syndrome of CRS in clinical practice,with remarkable curative effect.However,as the research on this prescription is mainly focused on quality control and composition identification,its mechanism of action is still unclear.In this study,systems pharmacology methods were used to carry out pharmacokinetic evaluation,drug targeting and pathway correlation analysis of TBF,so as to analyze the potential mechanism of this prescription on CRS.The main results are as follows: 1)At the molecular level,we obtained 85 compounds with high oral bioavailability and drug-likeness properties,among which angelica byakangelicol,kaempferol and luteolin are the typical pharmacodynamic molecules in Fructus Xanthii,Hedysarum Multijugum Maxim or Menthae Herba.2)Through target prediction and network topology analysis,195 targets were screened and found to be closely related to chronic rhinosinusitis.3)On the basis of clinical knowledge,combined with the mapping results of the target on the pathway,it is revealed that TBF exerts its therapeutic effect through the three modules of anti-inflammatory response,blocking virus infection and vasoconstrictor.Further in vitro experiments confirmed that the active ingredients of kaempferol and atractylenolide I significantly reduced the expression of inflammatory factors in mouse mononuclear macrophages,and verified the efficacy of the compounds from the perspective of anti-inflammatory.To sum up,this study focused on the multi-functional synergistic mechanism of TBF in the treatment of CRS.From the perspective of biological function of pharmacodynamics targets and pathway module analysis,this paper systematically analyzed the modular treatment mode of multi-component cooperation in the treatment of chronic inflammation by Chinese herbal medicines.It provides a precise strategy for the treatment of CRS and the research and development of new drugs,and also provides a new idea for the modern application of TCM.
