| With the change of times and the development of economy,the chemical industry has gradually shifted from mass production of basic chemical products to personalized and targeted special products.Drug production is a chemical product engineering with high cost,long time and high profit.At the same time,it has been gradually discovered that the treatment of many complex diseases requires multiple molecules acting on multiple targets for multiple regulatory purposes.Traditional Chinese medicine(TCM)has become a hot topic in the world because of its multi-molecular and multi-target characteristics.However,determining the mechanism of action between multiple components and multiple targets remains a huge challenge in the research of chemical products.The purpose of this study is to explore and explain the mechanism of Danzhixiaoyaosan in the treatment of depressive disorder by systems pharmacology.First of all,1064 kinds of active ingredients in Danzhixiaoyaosan were obtained by systems pharmacology technology,and a relatively complete database of compound ingredients of Danzhixiaoyaosan was established up to now.In addition,134 active components and 76 targets were screened out through oral bioavailability screening,drug-like evaluation and half-life screening.Then through the analysis of network pharmacology,the mechanism of Danzhixiaoyaosan in the treatment of depression was clarified.We found that herbal medicine mainly have an effect on depression by three functional modules,the inhibition of inflammation,neuroprotection and monoamine neurotransmitter.And the implementation of these functions depends on the operation of the complex biological pathway network,including six pathways in particular.This work not only provides an insight of the therapeutic mechanism of TCMs on treating depression from a multi-scale perspective,but also offer an efficient way for drug discovery and development from herbal medicine as complementary drugs. |
