The Animal Studies Of Biyan Tongqiao Spray In Treating Rhinitis

Biyan Tongqiao Spray is produced based on Chinese traditional medicine prescription of flos magnaliae powder(FMP) that including Ephedrae Herba Scutellariae, radix Menthae haplocalycis, herba Angelilae dahuricae, radix and Magnoliae flos.It has the effects of dispersing wind and clearing heat,relieving the stuffy nose. Biyan Tongqiao Spray has good treatment of allergic rhinitis, acute or chronic rhinitis. sinusitis and so on.Objective:According to the mainly treatments of Biyan Tongqiao Spray, a series of animal experiments are executed for discussing the safety and pharmacodynamics. Furthermore we make a further discussing on mechanism of action to lay a foundation for Clinical Application Reasonable,safety and effectiveness.Methods:1) Safety test:To determining the maximum quantity, observe the acu te toxicity of Biyan Tongqiao Spray. Throughing The nasal mucosa stimulatio n test to observe the nasal mucosa irritation of Biyan Tongqiao Spray.2) Main pharmacodynamics experiment:Pinna swelling modle on mice and granuloma m-odle on rats were used to observe the influence of Biyan Tongqiao Spray on a cute and chronic inflammation influence.Method of anti-bacterium in vivo was employed to observe the anti-bacterial action of Biyan Tongqiao Spray.3)Aller-gic rhinitis model on guinea pig was established to observe the behavioral cha-nge of allergic rhinitis model on guinea pig. HE dyeing method was used to o-bserve the nasal mucosa tissue pathology change. The level of SIgE and IL-4in serum of guinea pig were measured by method of ELISA. The expression of TNF-a、SP、VCAM-1、Bax in nasal mucosa were examined by using immu-nohistochemical staining method.We discussed the effect and the function mech anism of Biyan Tongqiao Spray in allergic rhinitis.Results:1) Acute toxicity tests results:the rat were treated with Biyan Tongqiao Spray and the maximum dose at12.82g(crude drug)/kg(equivalent to clinical employs660times as much as the doses),there was no toxicity and deaths in our study. This group of big rat did not appear toxicity reaction and deaths; Nasal mucosa excitant test results:After guinea pig treated with Biyan Tongqiao Spray0.77g(crude drug)/kg (equivalent to clinical decodes doses of40times), the animal was good in a condition and without abnormal in its appearance and behavior. There was neither erythema, edema in the localation, nor severely pathological change and inflammatory response in nose, and throat, the trachea and bronchi respiratory tract mucous membrane with pathological and histological examination.2)"Biyan Tongqiao Spray"could relieve the granuloma induced by plastic ring in rats and decrease the tumefaction in auricle induced by dimethyl benzene in mice.It also could protect the mice that it was infected by aureus staphylococcus and reduce the mortality rate.3) Biyan Tongqiao Spray could decrease the behaviour of allergic rhinitis,change the pathological symptoms of nasal mucosa.It also could decrease the level of SIgE and IL-4in blood.The expression of TNF-a、SP and VCAM-1in nasal mucosa were restrained,that of Bax was opposite.Conclusion:After animal with Biyan Tongqiao Spray12.82g(crude drug)/kg,th-ere was no obviously toxicity and deaths. After animal with Biyan Tongqiao Spray0.77g(crude drug)/kg,there was no obviously thrill.histologic examination has not seen the abnormal, and suggest that the doses is safe and reliable.Mai-n pharmacodynamics experiment results show that:"Biyan Tongqiao Spray"has action of anti-inflammatory, anti-bacterial and treatment of allergic rhinitis.The mechanism relates possibly to its effects of resisting the release of inflammator-y factor,modulating the immune Function, transfer of inflammatory cell and pr o-moting the apoptosis of inflammatory cell.
