Objective:A rat model of post-stroke depression was established to investigate the changes of serum norepinephrine,epinephrine,and dopamine levels in rats with post-stroke depression.Methods:1.The rats were randomly divided into two groups: 12 in the normal control group,12 in the stroke model group,12 in the depression model group,and 12 in the post-stroke depression group.Firstly,the middle cerebral artery occlusion model was established to establish the stroke group.The depression group model was established by the combination of chronic unpredictable mild stimulation and solitary culture.In the preparation of post-stroke depression model group,the middle cerebral artery occlusion model was first established,and the stroke rat model was successfully prepared,and then combined with the depressive model modeling method to perform the second model.Finally,a post-stroke depression rat model was obtained.2.Model evaluation criteria: The stroke model was assessed using the Longa 5 evaluation criteria;the depression model was assessed using body weight assessment,open-field trials,and sugar water consumption experiments.3.The contents of NE,E,and DA in serum of each group were detected by high performance liquid chromatography-fluorescence detection.Results:1.Behavioral and Body Quality Test Results:(1)Sugar water consumption test: Compared with the control group,stroke group,and depression group,the amount of drinking sugar drops in the post-stroke depression group(P<0.05).Compared with the control group,the consumption of sugar in the stroke group and depression group decreased(P<0.05).Compared with the depressed group,the consumption of sugar in the stroke group decreased(P<0.05).(2)Open-field test:(1)Horizontal exercise scores: Compared with the normal control group,the stroke group,and the depression group,the level of horizontal movement was reduced in the post-stroke depression group(P<0.05).Compared with the normal control group,the level of horizontal movement in the stroke group and depression group was decreased(P<0.05).Compared with the depressed group,the level of horizontal movement in the stroke group was decreased(P<0.05).(2)Upright exercise scores: Compared with the normal control group,the stroke group,and the depression group,the upright exercise scores was reduced in the post-stroke depression group(P<0.05).Compared with the normal control group,the upright exercise scores in the stroke group and depression group was decreased(P<0.05).Compared with the depressed group,the upright exercise scores in the stroke group was decreased(P<0.05).(3)Central grid stay time: Compared with control group,stroke group and depression group,post-stroke depression group had longer time stay in Central grid(P<0.05).Compared with the normal control group,the Central grid stay time in the stroke group and depression group was longer(P<0.05).Compared with the depressed group,the Central grid stay time in the stroke group was longer(P < 0.05).(4)Modified times: Compared with the normal control group,the stroke group,and the depression group,the modified times was reduced in the post-stroke depression group(P<0.05).Compared with the normal control group,the modified times in the stroke group and depression group was decreased(P<0.05).Compared with the depressed group,the modified times in the stroke group was decreased(P<0.05).(3)Body weight: The body weight of rats in the post-stroke depression group was significantly lower than that in the stroke group,the depression group,and the normal control group(P<0.05).Compared with the normal control group,the body weight of rats in stroke group and the depression group were significantly lower(P<0.05).Compared with the depressed group,the body weight of rats in the stroke group was lower(P<0.05).2.Biochemical test results:(1)Under the experimentally designed chromatographic conditions,NE,E,and DA in the sample serum were successfully separated and the peak shape was good.The standard curve shows a good linear relationship.Serum samples without other interferents.The experimental conditions can accurately determine the levels of NE,E and DA in rat serum.The peak times for NE,E,and DA are 2.96 min,4.08 min and 6.53 min.The other impurities in the serum sample are well separated from the peak of the analyte.(2)The levels of NE,E and DA in the post-stroke depression group were significantly lower than those in the normal control group,and the difference was statistically significant(P<0.01).Compared with the stroke group,the levels of NE,E and DA in the post-stroke depression group were significantly lower,and the difference was statistically significant(P<0.01).Compared with the depression group,The levels of NE,E and DA in the post-stroke depression group were significantly lower,and the difference was statistically significant(P<0.05).There was no statistically significant difference between the stroke group and the depression group(P>0.05).Conclusion:1.The levels of serum NE,E,and DA in the post-stroke depression rat model were significantly decreased,which may be related to the pathogenesis of post-stroke depression.2.The decrease in the level of catecholamine neurotransmitters is highly correlated with the degree of ethological changes in rats.
