| Objective:To observe the effect of modified bushen recipe on the expression of serum RLP-C,ApoA1,ApoB and uterine ER in rats with POF hyperlipidemia,Serum RLP-C was used as the intervention point of this prescription on the rat model of premature aging HLD.Methods:After observing the estrus changes of SD female rats for 4 ~ 5 days,48 rats with normal cycle were selected and randomly divided into blank group(12 rats)and POF group(36 rats).The POF model rats were injected intraperitoneally(Ip)by cyclophosphamide and Busulfan(BUS),and the rats in the model group were fed a high fat diet,while the blank group Ip Saline was still fed basic feed;After the POF MODEL was established successfully,the model group was randomly divided into four groups: Chinese Medicine Group,Western Medicine Group and POF Model Control Group,each with 12 rats.Each group was treated with the corresponding solution,the blank group and the model group were filled with 0.9% NS,the traditional Chinese Medicine Group and the Western medicine group were filled with kidney-tonifying prescription liquid and Estradiol valerate suspension respectively,and the time of administration was 20 days.Daily observation indicators are: 1.Rats in general,2.Changes in the estrous cycle.Rats were sacrificed at intervals of the cycle,blood was collected,ovarian and uterine tissues were taken out,and the morphology of liver was observed.Serum FSH,E2,RLP-C,ApoA1 and ApoB were detected by Elise method Immunohistochemical method was used to detect ER level in uterus,and the results were analyzed statistically.Results:1.Weight comparison of rats at different stages:There was no significant difference in body weight between the blank group and the POF Model Group(P>0.05).The body weight of the Blank Group was higher than that of the model group(P<0.05);After administration,Day 1: The Body Weight of Blank Group was higher than that of TCM Group,Western Medicine Group and Model Group(P<0.05),Day 9: The Body Weight of Blank Group was higher than that of model group and Western Medicine Group(P<0.05),Day 18: There was no significant difference in body weight between the four groups(P<0.05).2.The changes of serum FSH and E2 levels in rats: On the 3rd day after CTX + BUS model making,FSH and E2 in model group and blank group were significantly higher than that in control group(P<0.001),and E2 was significantly lower than that in control group(P<0.001);After 20 days of drug intervention,FSH and E2 increased and decreased in the model group(P<0.05),Compared with the model control group,the FSH content of the two groups decreased significantly and the serum E2 content increased(P<0.05),the FSH content of the two groups had no statistical significance,but the E2 content of the TCM Group was slightly different(P<0.05).3.The weight of rats before and after the MODEL was established: Before the model was established,there was no significant difference between the four groups(P>0.05).The body weight of blank group was higher than that of model group(divided into Chinese,western and Model Control Group)(P<0.05).4.The levels of serum APOB and RLP-C in model group increased,while serum APOA1 decreased,which was significantly different from that in normal group(P<0.05).After treatment,the levels of serum APOB and RLP-C in traditional Chinese medicine group and Western medicine group were significantly lower than those in model control group(P<0.05).5.The expression of ER in rat uterus was significantly lower in model group than that in normal group(P<0.01)and increased in model group(P<0.05).Concluston:1.POF MODEL was established by CTX + BUS,the body weight of rats decreased obviously,until the 4th day after CTX + BUS model.There was no difference in body weight between TCM Group and Blank Group after 9 days,and the recovery speed was the fastest.2.By increasing serum E2 and decreasing serum FSH level,bushen prescription and Estradiol valerate can effectively treat POF,and the effect of traditional Chinese medicine on increasing estrogen level is better.3.By lowering the level of serum APOB and increasing the level of RLP-C and APOA1,bushen recipe and estrogen are effective in regulating the disorder of Lipid metabolism of POF.4.In the experiment,Chinese and Western medicine increased the expression of ER in uterus of POF rats,and the expression of ER in epithelial cells was the best,interstitial cells was the second.5.Bu Shen Decoction increased the expression of ER in uterus in HLD related dyslipidemia of POF,which was related to RLP-C. |
